I have literally done a flight from EGCC-WSSS-YPPH (using the 747-400 Polar DHL) in one go, though I did have a tad bit of a break in WSSS. that was around 20 hours. I too have gone around the world going in the following order EGLL-KJFK-KLAX-PHNL-NZAA-WSSS-HKJK-ORBI-EGLL (using the 787-9 British Airways)

Absolutly great flight, a little suggestion which is a very helpful thing, a little button on the center panel, its called auto pilot which maintains altitude, speed, navigation, etc. it can also do an auto landing, but it was a great flight

As a sydneysider, I can confirm he did force the airport to close for a few hours after that Ryanair landing

So for a 21 hour flight there would be 4 pilots in the cockpit. So they could switch and the pilots could get rest
