What is actually the difference between Fusion method A and Fusion method B? Is there any rational as to which method is to be prefered depending on the individual circumstances?

the vigorous shaking interferes with fusion. I do not believe this protocol is correct

Are you referring to the step in which the fusion is put into the semi-solid Medium D for HAT selection prior to plating? Since Medium D is a semi-solid media and much more viscous than a typical liquid media, shaking is required to mix the sample. In addition, the force of the shaking on the fused cells will not be as physically vigorous (violent) on the cells as they are in semi-solid media. As well, our protocol includes a recovery step following fusion and before plating in selection media. We can address your question in further detail if you email techsupport@stemcell.com.
