Dr. Jim Allison was deeply influenced by his days at UT and the music scene in Austin in the 1960s and
70s. He recounts his progression driven by curiosity as an undergraduate student and laboratory dishwasher, followed by graduate studies that led him to develop an unrelenting desire to understand the workings of the immune system, particularly T-cells. He identified the molecules that provide the fundamental regulation of T cells, including the ignition switch (antigen receptor), gas pedal (CD28), and brakes (CTLA-4). These findings led Allison to develop immune checkpoint therapy as s way to unleash T cells to attack cancer cells, which can lead to cures for many different types of cancer. He shares some of his adventures as both a scientist and a musician.
Dr. James Allison is Regental Professor and Chair of the Department of Immunology, the Olga Keith Wiess Distinguished University Chair for Cancer Research, Director of the Parker Institute for Cancer Research, and the Executive Director of the Immunotherapy Platform at MD Anderson Cancer Center. He has spent a distinguished career studying the regulation of T cell responses and developing strategies for cancer immunotherapy. He earned the 2018 Nobel Prize in Physiology or Medicine, which he shared with Dr. Tasuku Honjo, "for their discovery of cancer therapy by inhibition of negative immune regulation." Among his most notable discoveries are the determination of the T cell receptor structure and that CD28 is the major costimulatory molecule that allows full activation of naïve T cells and prevents anergy in T cell clones. His lab resolved a major controversy by demonstrating that CTLA-4 inhibits T-cell activation by opposing CD28-mediated costimulation and that blockade of CTLA-4 could enhance T cell responses, leading to tumor rejection in animal models. This finding and a great deal of persistence paved the way for the field of immune checkpoint blockade therapy for cancer. Work in his lab led to the development of ipilimumab, an antibody to human CTLA-4 and the first immune checkpoint blockade therapy approved by the FDA. Among many honors, he is a member of the National Academies of Science and Medicine and received the Lasker-Debakey Clinical Medical Research award in 2015. His current work seeks to improve immune checkpoint blockade therapies currently used by our clinicians and identify new targets to unleash the immune system in order to eradicate cancer. 2012 - I had melanoma in the brain, both lungs, liver, kidney, gall bladder and spinal cord. I was so fortunate to be placed in the first clinical trial of ipilimumab and nivolumab at Yale - New Haven Hospital. After two craniotomy, three Gamma Knife radiations and the clinical trial, I was free of cancer after eight months. Ten years later, still cancer free, many thanks Dr. Allison and Dr. Honjo. Side note: Dr. Allison was asked, which was more exciting, meeting the Pope or playing with Willie Nelson. Dr. Allison’s reply was that it wasn’t a fair question.

This man has saved and is saving many lives with his knowledge and passion. My daughter is at MD Anderson benefiting from his developements on the Cancer front on this very morning. Thank You Mr. Jim!!!

Velvett, I wish the absolute best for your daughter and you. When your daughter experiences side effects, please have her report them to her on oncology team. There are treatments to counter the side effects. Please read my post on this page.

I have renal cell carcinoma in my lungs. I’m having great results with daily oral chemo and immunotherapy every 3 weeks for almost a year. My oncologist studied under him at MD Anderson. Thank you Dr. Allison!

I've been doing a ton of research on various cancer treatment options and focusing on treatments like this. My dad has been dealing with multiple myeloma for 13 years and I refuse to accept it can't be cured. Keep up the good work!!

After dealing with melanoma and MPN, I am truly amazed by healers that can dissect these deviant flaws and cure so many of us with their grit and tenacity! May all of us mortals attempt to improve our space with our tools of life. These cosmic MD's rock!+++

Amazing work. This how medicine should be, is getting rid of cancers at any stage and saving people lives. Keep working hard to find a cure. :)
