This is my own analysis but Namju or 남주 is short for 남자 주인공, a noun which means male lead, Yeo Juda or 여주다 is short for a whole sentence, 여자 주인공이다 which means "it's the female lead"

But i hope at least you understand the pun hidden in these names (in fact i figured out almost every character of this movie has pun in their name, even the extras, but it would be too long to explain :)))
