This video is the third part of the lecture on flow cytometry and hematopoiesis. In today’s video of the flow cytometry series, we will be covering step by step journey of B-cell hematopoiesis and maturation with changes in CD markers and also the origin of all major B-cell neoplasms including B Lymphoma and acute lymphoblastic leukemia.

If you have missed our previous videos on the basics of flow cytometry where in part 1 we discussed working principles of the flow cytometry machine, interpretation of graphs, In part 2 of the flow cytometry series, we understood what CD markers are, and how hematopoiesis and cd markers lay down the foundation for understanding flow cytometry and interpretation of flow cytometry plots. We also learned cd markers of myeloid series on flow cytometry plots. If you have missed our previous two videos, there is a link you can see up, do watch them for having a better understanding of this flow cytometry lecture on hematopoiesis and B cell development and in no time, you can become a champion in the interpretation of flow cytometry
B-cell neoplasms are clonal tumors of mature and immature B cells at various stages of differentiation. Each b lymphoma and acute lymphoblastic leukemia subtype bears a phenotypic resemblance to B cell at a stage of differentiation during the hematopoiesis.
So instead of mugging different cd markers for b lymphoma and acute lymphoblastic leukemia, if we just remember the normal postulated counterpart from which b lymphoma and acute lymphoblastic leukemia arises, flow cytometry plots will NEVER BE A BRAIN TEASER AGAIN.
Now let’s move to the part we all want to learn- that is diagnosing b lymphoma on flow cytometry. Diagnosing b lymphoma and acute lymphoblastic leukemia by flow cytometry is a challenge. Let’s understand why?
Many a times in flow cytometry we get a sample from a lymph node or a peripheral blood to be tested for presence of lymphoma . However lymph node might be having reactive lymphoid cell proliferation due to infection , or peripheral blood lymphocytosis may again be reactive . So how do we differentiate both the conditions on flow cytometry plots? The answer is to learn the must know CD markers!!
Each lymphoma subtype bears a phenotypic resemblance to B cells at a particular stage of differentiation. So instead of mugging different markers for lymphomas , if we just remember the normal postulated counterpart from which lymphoma arises , identifying LYMPHOMAS on flow cytometry WOULD NEVER BE A BRAIN TEASER AGAIN.
So that’s all for today in flow cytometry. I think it would be very helpful if you could do a tutorial showing actual flow charts illustrating specific conditions like various lymphomas, leukemias and other lymphoproliferative disorders. You could explain what exactly are the features that allows a hematologist to diagnose a specific condition by flow cytometry charts. I would like to add that all your previous videos have been excellent and congrtgulate you on doing an outstanding job. Do keep it up.

Am very late to this discussion but came across it as I was looking for an explanation in a case I have where CD4 count is .10 CD8 is .24 and CD3 is .66 (all 10*9/L). What I cant figure out is CD4 added to CD8 would only be .34 which makes CD3 194% of that. I thought Cd4 plus Cd8 should almost equal CD3. So, what then is expressing CD3 but is not a CD4 or CD4?

on the antigen independent phase of maturation you mentioned that the B cell pan markers are (CD19, CD22 and cytoplasmic) CD79a. all of the sudden on the antigen dependent phase you replaced CD22 with CD20 as a pan marker for B cell. Is this accurate? it is confusing, you didn't mention CD20 at all before this stage. Please clarify.

really great video... will be highly obliged if make somthing on PNH,DHR and EMA dyebinding in flow cytometry i do procedure in my department but principles are not very clear..
