The fact is at a particular position of the scanning mirror or scan time, one particular wavelength gets into constructive interference giving rise max intensity at that wavelength.

@@devanggondaleeya4321 Certain peaks at a certain frequency indicates a specific functional group, you can find out what functional group your sample have by matching FTIR spectrum to a library.

This is all very clear. Thank you. Just one question. If using two different bacteria to test for biodegradation, can the peaks differ depending of the organism used? The peaks i got after incubation with Pseudomonas have a lower absorbance than the peaks obtained using ideonella. The compound is neat PET.

As IR shows weak absorption compared with UV-VISIBLE radiation, generally transmittance is plotted on Y-axis.
