The prank is that SHIOZUKE sounds similar to SHIZUOKA. Polka was saying a lot of district/city names ( hence the outfit changes) forcing Miko, midst hype to say "SHIZUEKE" a mix of the two words. Classic ringmaster act, from Polka.

Polka is saying the names of prefectures and then dressing up as things the prefectures are famous for. Nara has deer that roam in city parks, Hokkaido has lavender and crabs, Aomori is the Nebuta Matsuri, Akita is the Kanto Matsuri, Okinawa is a hanagasa hat, and Shizuoka is a tea farmer.

I find it so interesting that this sounds more similar to people who speak Japanese than English. As someone who only speaks English and doesn't know what the words actually mean, they sound fairly distinct, but I suppose to a Japanese speaker, it's not as much. I know that generally makes sense, but considering it's more about the sound the words make than words themselves when it comes to tongue twisters, I just find it interesting. Or I'm just looking too deep and Miko is just super ELITE

if you split the words into their japanese syllables (since at the base form they will be thinking in syllables instead of an alphabet), you have shi-o-zu-ke and shi-zu-o-ka. So sure, they're clearly different words, but I hope it's easy to see how you can accidentally swap the syllables around.

At the end of the video after the subs stopped, Polka said "That was super clean. What an elite way to be fooled" That's exactly right. Miko's ability to not only get bamboozled but to fully bring out the bamboozledness of the prank is truly a talent.
