I was filming.
This was a research project I did with a friend when I was a sophomore in college, so it was from a few years ago. We made an algae bioreactor out of 2L coke bottles, sterilized them with alcohol, then put in sterile salt water growth media and inoculated with some nanochloropsis we got as a sample from a marine bio lab.
We had it on a light cycle and always aerating through a series of tubes we hooked up to sterile pipettes, capped with sterile cotton plugs.
Once the growth was dense enough, we tried to figure out how to best filter the algae, and read that NaOH caused aggregation of the cells. We tried that and it worked, made the clumps easier to filter.
After we recovered our algae, we macerated it in liquid nitrogen and sand and did a sohxlet extraction on it.
I will put together that footage now, it's kind of scattered and old but it was a neat proof of concept. A lot of work for not a lot of oil in the end, but was a good learning experience.

Do you have any more content on this? Or even recommended resources. I haven't seen Sohxlet extraction used. I've mostly seen transesterification so I'm curious how you came to this process.

So you basically fermented water and algae to get alcohol, tgen separated the alchol from the glycine. Could just distill wine or beer ?

will using recycled PET bottles like this be suitable for human-grade food algae like spirulina? or will it degrade overtime and become unsuitable for food production?

I was wondering which is less energy intensive; Producing sodium hydroxide for flocculation, or centrifugal separation?

+Keith Reynolds I would say centrifugal. If the machine is built proper it should be efficient and long lasting and possibly also fully recyclable later on. It could also be powered via renewable means such as leg/arm power or solar, wind etc. Possibly even being powered by the oil made from the system itself.

Does the sodium hydroxide kill the algae and will sodium hydroxide kill spirulina? I was also wondering if you knew of a way to cleanse spirulina of unwanted bacteria and keep it alive in a paste form?

You can't cleanse out the bacteria and keep the algae without something like an antibiotic solution, but that's not needed. Most algae culture is grown under sterile conditions, ours included to the best of our ability. You can filter it and store it as a paste. It's very common to do that for use in aquaculture feed, etc.

He did say it was a 1 micron Sock filter, Try an aquarium supply house. Saltwater usually has fancier stuff. Lol, If you're still looking into this a year later!

We ended up getting these online, I think they were used in bio-diesel production? I believe 1 or 2 micron size. This was a long time ago.

Nothing added, just a synthetic seawater mix from a fish store. The algae photosynthesize for energy so no additional compounds needed.

what exacty did you use as a pump for the air

Just an aquarium pump for $5, pumping air through tubes we pre-sanitized with dilute bleach, and we used large sterile plastic/glass pipettes with sterile cotton in the top to aerate from the bottom of the bottles.

you need organic solvents to extract the oil out of microalgae such as hexane or methanol or ethanol, not NaOH, NaOH is used as a catalyst in the reaction, not the solvent.
