Hi Miss, thank you for your tips! Is it possible if you could go over "preparing a coherent and well-written essay", on tips on linking and connecting ideas/paragraphs in the biological essay as well as knowing what counts as waffle and what doesn't count as waffle in your explanations as these are the main killers of the biological essay? My own Model Answers: 1. Transmission electron microscopes work by using a high energy electron beam, focusing this onto the sample, using electromagnets. Denser areas of the specimen will absorb more electrons, appearing darker on the image. Optical microscopes work by using light to view an image, focusing it using a glass lens, they have a shorter wavelength and so optical microscopes have a lower resolution than TEMs because of this. Limitations to a transmission electron microscope is that although it has a high resolution and can view the internal structures of a cell (such as mitochondria, ribosomes, endoplasmic reticulum) it cannot view living specimens as the sample must be viewed in a vacuum. A vacuum is needed to remove air, as air can absorb electrons and distort the image. TEMs can only observe 2D monochromatic images,and they have a complex staining process in which can increase the likelihood of artifacts being produced. Limitations to a optical microscope is that is has a lower resolution due to the longer wavelength and so cannot view all the internal structures of organelles in a cell. Although it can view living specimens and it can view them in colour. 2. DNA is made up of monomers called nucleotides. They are made from a deoxyribose sugar, a phosphate group and a nitrogenous base (either adenine, guanine, cytosine or thymine). These nucleotides form phosphodiester bonds through condensation reactions between the deoxyribose pentose sugar and a phosphate group on another nucleotide. The polymer form of DNA is coiled into a double helix consisting of two separate and complementary strands. Between the strands there is complementary base pairing through hydrogen bonding. Adenine bonds to thymine with 2 hydrogen bonds, cytosine to guanine with 3 hydrogen bonds. The phoshodiester bonds formed between the nucleotides create a phosphodiester backbone which holds the DNA nucleotides together. These strands of DNA are wound around proteins called histones in eukaryotic organisms that condense to create chromosomes.
