Dear Scientists,
LAMP, short for loop-mediated isothermal amplification, is an alternative technique/method for DNA amplification and has some advantages over PCR. The principle of the LAMP assay is based on a selection of 4 to 6 primers as shown as in this animated video*.

How is each amplicon produced? How is the strand displacement accomplished, is it a function of the polymerase? Why is the loop formation necessary?

How both DNA denaturation and primer binding is occuring at same temperature?As both of them are opposite reaction?

Heyy, I did LAMP today. The experimental setup was pretty straightforward. BUT, trying to understand the principles behind it, as concisely explained in your video is quite a challenge for me to wrap my head around it. I'll have to do more reading for sure.. 😂 Thanks for the explanation!

Excellent! Finally, I understood the loop structure formation. However, how does it makes the DNA duplication faster?

In PCR there are 3 cycles which combined take up some time to generate the daughter strands. In LAMP the synthesis continues constantly.. further: using 6 primers in case of LAMP, provides more contacts for polymerase recruitment!

The animation is very helpful. It will be even better if was shown at least one cycle after the annealing of external primers, indicated in 3:14. Unfortunately, I live in Brazil and my financial situation is not so good as I would like, but your animations deserve much more than $ 2.00. And thank you for answering my questions!

I read that most DNA denatured at 75 degrees, does the DNA denatured at 60-65 degrees in LAMP or just some location? Can this become a limitation of LAMP?
