Meselson and Stahl experiment - This lecture explains about the famous meselson and stahl experiment that explains the mode of DNA replication. Meselson and stahl did the experiment to prove the semiconservative model of DNA replication. The experiment is performed by growing e. Coli bacteria in heavy nitrogen medium and then transfered to the light nitrogen medium to check the incorporation of new light nitrogen to know the model of semiconservative DNA replication.

There's a mistake- after the first round of replication, conservative was ruled out, because conservative replication would not produce an intermediate strand... it would be light or intermediate, never heavy. The second round of testing was needed to distinguish between semiC and disp. models. Then it was discovered that only two gradients ever appeared; the medium and light. Dispersion would have multiple variants present based on its more random nature.

Sir what if we took E.coli containing normal nitrogen initially and then provide a medium of heavy nitrogen for 1st ,2nd ...and so on generations , will we get the same Conclusion of semiconservative nature of DNA ??? Also after 1st generation , only medium density DNA should come in centrifugation and in 2nd generation only medium and light bands should come as in ncert also .....😊😊😊
