Sure can. Just adjust the baking time. For cupcakes, around 18-20 mins at 180°C

No, because only buttermilk will give you a fluffy texture.

You can add 1tbsp of vinegar to your milk or 1 tbsp of lemon juice mix and wait a few minutes and it will have acidic taste like buttermilk
