Duuuude!!! Just struck GOLD finding this video!!! Been struggling with bacterial riders for a whiiiile now. Basically have narrowed it down to my LFH filter been saturated, and every "open jar" agar/grain transfer I do in front of it gets bacteria that I later transfer to tubs and get Trich. Really frustrating cuz the myc looks "OK" but at the end of the cycle, all that effort and resources are lost. But, looking at your video I've learned that they may ALSO be embedded bacteria coming in the genetics purchased. 🤔 Cheers man! Keep doing these videos! One about possible origins of such contams/bacteria would be nice!!!

Ah man, My first grow I did 10 jars and I didn’t get one that was completely clean. I was able to salvage enough to do one tub of gt And it worked. Got a bunch of really small ones the first flush, I was so happy it worked. It was mss straight to grain and I wasn’t very good at hydrating the popcorn at all. Plus I don’t think I did even a full 90 mins.

That's a phenomenal idea actually. Smart. The mycelium will reach across while the bacteria is held on the contaminated side 🎉🎉🎉🎉
@MCH400
@MCH400 8 күн бұрын
@@Mycophiliahe means embedded bacteria. Basically youd leave 1/3 of the dish with the bacteria embedded mycellium then empty the rest out and fill with fresh agar alongside that wedge and then cut a "ditch" out of the agar so there is a line of clear space between what's already colonized with the bacterial mycelium and the side with the fresh agar. The mycelium will walk over that gap but the bacteria in theory wouldn't transfer along with it. I haven't personally had to do this, but it certainly does make a lot of sense

Thanks for this content, its hard to find information on hidden contamination, but it's quite common for me! One idea I wanna share with you @Mycophilia: I remember Mycoplasma are smaller then other bacteria and prefer to stay intracellular, not intercellular. Maybe they are your hidden embedded contamination. Did you try growing this kind of contaminated mycelium on Agar-water? all the best!

Certainly could be the case. As for water agar, yes, and ime it does nothing for embedded contams unfortunately

Trich kinda takes over in growth quickly, if it's spotting light green it's penicillium, trich spreads much faster and has a forest green color when matured, penicillium takes longer so you can let it go a little longer before you have to toss it out, gives you chance to get some fruits out if you're at that point that is. Love the vids brotha!!! Keep fighting the good fight!!! 🦍🙏💪🏿

I loved the video, but I am not really sure what to look for. I see the little yellow metabolites, and the fluffy white jars definitely look strange. Very interesting, but what makes you think the jars with the white mycelium are contaminated with bacteria, or even contaminated at all? Have you done some tests that support that notion? Might not something else actually be going on? (BTW, next time why don't you slice the white mycelial mass into 1.5-2 cm 'breadslices' with a knife and try putting them btw a thin 50-50 CC-Verm bottom and thicker 50-50 CC-verm top layer and see what happens. I suspect breaking up spawn that looks like that might not work very well.)
@Mycophilia
@Mycophilia Жыл бұрын
They are 💯 contaminated, once you’ve grown for some time you know what’s contaminated and what’s not. I have listed a number of reasons on this video, the primary ones being the extremely thick growth that makes it impossible to break up, along with all previous attempts with this culture always ending up with the same green mold, at least those that have been spawned to bulk. The ones that have not (bottle fruited), have not fruited in 2+ months of fruiting conditions, in addition to growing very strange mycelium masses after the first month, a hallmark of contamination. As for your idea of slicing, a better method exists called jar/top fruiting. It is better because you don’t need to break up the mycelium at all. Hope that helps.

If your bulk is going bad then you have 1) Not fully sterilized (longer higher temps required) Why does this happen???? Elevation has an effect on temp. 2) You introduced a contaminant (Lysol/alcohol and draft hood) 3) Your inoculate is contaminated (do a better job on sterilizing and use smaller wide mouth jars)

Is there a chance it's fine? I feel like a bad parent. These are my babies 😢

After scaling up my grow I got a lot of bacterial contam, still trying to rule it out. I was using popcorn, never had a problem before while using jars, but when I moved to bags I got into problems, all of my contams are bacterial, not a single colorful mold :( Now im using millet, perhaps it will be easier to achieve clean spawn.

Im curious why don't you use water agar to isolate the Mycelium from the bacteria ???
@Mycophilia
@Mycophilia Жыл бұрын
Because water agar does nothing to embedded contams as the mycelium itself is contaminated, it’s not something you can easily isolate like regular bacteria via water agar.
@seanhenderson1522
@seanhenderson1522 Жыл бұрын
This would be the first I have heard of “embedded” bacteria that can not be removed. Can you kill it with antibiotics? It seems like you’re suggesting that there is a bacteria that is living and feeding from the mycelium it’s self, definitely seems plausible.

Great video! Will keeping my grains at a slightly cooler temp help deter the bacterial growth and allow the mycelium to overtake it?
@Mycophilia
@Mycophilia Жыл бұрын
It’ll help deter bacterial growth, but if the bacteria’s already ‘triggered’ then it won’t make much difference as the mycelium will always lose eventually

I had a box that formed hard nodes all over the surface and wouldn't fruit, never seen something like that before, smelt great tho even when I broke it up and threw it away, wonder if it was some sort of embedded bacteria, I got rid of all the grain cause I tried to fruit it 4 times and gave it a month each time no success

Question, were those white jars of the compressed myc started from what looked like a singular culture on an agar plate from spores? I think people get germinating spores mixed up at times. Spores can germinate and grow by themselves. They only need to meet up later with other germinated spores to go on and grow healthy mycelium and eventually fruit. This may be the problem with that myc. I'd try introducing another sample of myc that came from the same strain and see of they marry 🤷

have you added ashes just wanting to know if your hypothesis works ? i personally wanna try those jelly-looking colored balls that you soak in water till they become the size of a marble after 24 hrs later. people use them inplace of soil to grow most houseplants anyway i was thinking just adding a few dry balls when they are the same size of rice and it works just like white rice to suck the moisture out of your cell phone. like a silica packet does.idk ill let you know how it works 🙂

@@Lexington-Felix orbees only hold moisture, and you have to use nutrients to keep a plant alive. Along with frequent replacement of the spheres of polymer and water. Mushrooms are very hard to grow outside of their natural environment. In nature they can survive other molds and bacteria. In the lab they require a sterile environment.

Hey! I think I'm dealing with the same bacteria as you. I've also done multiple transfers to both agar and grain and still am seeing those big fluffy clumps on top. Do you know if it's definitely bacterial, or could it be a mold or yeast? If it's bacterial then perhaps a transfer to antibiotic agar could clean it up?
@Mycophilia
@Mycophilia Жыл бұрын
Definitely worth a try. I would recommend doing at 2-3 xfers on antibiotic agar, and then 2 xfers on regular agar to make sure it won’t come back. This is because bacteria will always eventually outsmart the antibiotics eventually

Hey I had a grain bag w some wet rot and a lot of v healthy mycelium. I mixed it into substrate and it did smell for a bit but now the smell is gone and the cake looks amazing. Should I be worried about the fruits having some toxins from the wet rot? Can the mycelium eat the bacillus and any toxins they would have thrown? Any tips on what to do w the fruits? I have a feeling they are going to “look” v healthy and don’t want to waste the bag I’ve had a hard time salvaging this strain! Thank u!!!

Apologies if you've already addressed this in a previous video, but when fruiting truffle producers, will the fruits be more potent the longer i let the spawn colonize? Or as soon as the jars is colonized(little to no truffles forming) spawning to fruit will be the same potency?

the fruits are tiny and potent as it is. Recent run was 6 months truffle then fruiting trays with peatmoss casing, but I'm going straight fruiting for Tampa / Mexi this round. Will see if they are in the same ballpark.

For some reason I have better luck with rye berries than brown rice, and I've tried the 90 second uncle Ben tek and no luck! Ive just started out and did as much of the sterilisation I could with everything. I did an oyster batch but looked like white fuzzy mold starting after a few days so got rid of it also those random white dots in various places. Then I redid the process trying to be even more careful with another LC syringe (GT) I got in a pack. Was as sterile as i could be with alcohol, etc but on rye and oven baked at 80 deg C for about 2.5 hrs since i dont have a PC yet. Made sure to squirt down side to monitor, after 3 days I can see white either whispy or early signs of white fuzz. Should i leave it and monitor or perhaps toss it while its pretty early? Appreciate the content and learning a ton so far.

I’m having that issue with some strains too; clean growth on petri but contam when I transfert them. I noticed a very very thin growth underlying the mycelium on petri and it’s always growing just a little bit ahead of mycelium The growth pattern is more chaotic than mycelium and I need to look very close with bright light to see it. I think it’s parasitic. I wish I could identify, all I know is that it’s probably not gram negative, as I tried to transfert from petri to LC with high dose of gentamicin.

I started doing this chasing mycelium into fruit about a year ago and I use the uncle Ben’s method but the last six syringes I received all turned into super green nasty. My guess is that it’s Trichoderma has a very foul smell to it. my first I’d say eight times was very successful. I have not changed my procedures if anything I have gotten even more excessive compulsive about cleanliness. I am guessing that I am getting contaminated syringes. From your experience, is that a possibility?
