Sir, if we take one DNA molecule and attach the Taqman probe to one strand, then how the probe will increase its fluorescence, as DNA will replicate and produces multiple copies.

RT-PCR is a manufacturing process NOT a diagnostic tool. RT-PCR amplifies the target so that one can do further study; it does not and cannot distinguish between something that is actively infective or not infective.
