Ferns are epiphytes and are widely cultivated for ornamental purposes. It is undoubtedly a great choice for a house plant.
The spores are often found underneath the leaves, which are protected in a capsule called sorus.
Such propagation method is much easier to be grown outside a lab as the spores are powdery to be disinfected via surface sterilization.
A successful plant tissue culture protocol was conducted and there was no significant difference between the surfactants using Plant Preservative Mixture, PPM and Clorox.
A germinated spore produced a structure called prothallus, which is a love-shape structure that is green in colour.

is this last medium with nutrients? i mean water in last jar u thrown spores in? u used ppm vs chlorox ?

@@jedz86 yes, it’s MS basal media. At this stage, we usually don’t add any hormones as there might be contamination. Moreover, the growth is super fast once it’s germinated without contamination.

Hi im trying to replicate your methods but some details arent clear. Based on the comments I've understood that you used 1-10% or 5-10% clorox solutions for 30 minutes and one rinse with distilled water before culturing on the plates. Unclear details are: What is the concentration of PPM used ? do you add it only to the first sterilizing solution or do you also add it to the culture medium? How long did you have them running under the tap water? Did you have them in the same tap water when u started in the bench? Did you go straight from the tap water to the clorox solution? After the clorox solution, did you only rinse the spores once with sterile water? or 3 times like most tissue culture protocols? and just curious what was your contamination rate? I tried it already twice and i have almost 100% contamination. Any help is appreciated thanks.

1. PPM is strictly depends on the conditions of the explants (spores in this case). You may use 5, 10, 15, and 20 mL per liter in the gelled media as I’m not sure if the growing condition of the plant bearing spore is “clean” or not. 2. More than an hour. 3. Yes, same. 4. Quick dip in 70% ethanol before Clorox. The concentration of Clorox is strongly dependent on the amount of the spores per tea bag and the penetration of the tea bag. 5. All sterilization process in my videos are: Tap water> 70% ethanol> Clorox> rinsing with sterile water. The reason I don’t put concentration used and duration is because everything has to be adjusted based on the explants we used and the status of the explants.

That was great thankyou! Can you tell me what solution the spores were pipetted into the Petri? It looked like it set in the dish or was there medium in the dishes already?

It was the sterile distilled water from the rinsing part of the sterilization..

In the last slides of the grown out Petri dishes, have they got ms/agar solution media and the explants have been transferred to these or are they just grown out to the size of the last slides in the water?

Sorry if I am confused. In the very last slide with most growth it looks like they are on agar? All the rest look like water.

Hi. Im currently busy inoculating stag horn fern spores. I was wondering what percentage dilution of your disinfectant did you use? How long did you leave the spores in the disinfectant? What yellow liquid did you add before you disinfested the spores? What agar media did you use? Love the video!

5-10% bleach solution... The duration varies according to the amount of spores used per batch... Roughly 30-60 mins

@@carlschulenburg9032 normal medium is fine...I used MS medium without any PGR for all videos...rinsing can be many times depends on your trials... Pipette of any brands is the same...just that you need to autoclave the tips before you use them...

@@hholi Hi, actually every medium works, just that MS medium is the most easily available and ready for the stock preparation…there are many cases where you can see people adjust the macronutrient as MS is well-known as high salt concentrations…

How many months do the spores take to germinate? I tried to germinate mine, but all I got is some tiny white fuzy colony but its only been one month. Confident not fungus since it didn't spread to the whole petri plates. Please help. Thank you!

You may need to add some drops of water to make sure they are fertilized…whole process would take about 1-2 months depends on the species

You need some moisture to make sure it’s fertilized between individuals… I didn’t proceed with further stage as this is the stage that client wants as they mentioned that induction of thallus phase in nature is very slow

You should not use gloves while burning with alcohol in the cabinet. It may have bad consequences ..!

Thanks for reminding, but, alcohol itself is a dehydrating agent...it will dehydrate your hands...one must be always aware of the alcohol on the gloves before it dried up...I've been wearing gloves for 5 years working on tissue culture and it's not an issue for me...for beginner, one must have extra precautions on it...
