Preparation of MS media for tissue culture

For tissue culture we need a medium for our explant to grow . A medium which contains all nutrients (organic and inorganic nutrients), amino acids and vitamins so that explant is cultured and grown in-vitro condition.
Most of the times we use ms medium(Murashige and skoog medium).
The one we prepared and displayed in video.

Maam where is your macro elements. You mentioned micro element stock instead of macro element stock. One is important thing is that you should add the stock in decreasing the quantity means firstly add macro then micro after that trace and remaining. The most important thing which you forgotten is that you not mentioned the measuring of pH. Without the pH measurement your media could not be solidify.

Inorganic salts stock which i mentioned have macro elements in it. We prepare macro element stock separate and micro element stock separately and add required ml of each for ms media preparation. And clearly I have mentioned the ph in the chart at the begining i.e. ph = 5.8

This was by far the best video,no other channel explained the steps for preparing exactly 1L of MS
