2024 Introduction to Biophysical Techniques
In this video, Poppy Llowarch and Laura Usselmann give an overview of high throughput screening at AstraZeneca. In first part of the talk, they explain how to set up dye-based differential scanning fluorescence (DSF) assays. They explain how they can used to prosecute purified protein targets that would be otherwise intractable. They detail the conditions needed for optimal results and how to analyse the resulting data, giving examples for hit compounds. They also describe a label-free nanoDSF method. In the later part of the talk, they cover a cellular thermal shift assay (CETSA). They describe how they set up an isothermal method in combination with a NanoLuciferase output to give optimal results, giving an example of a successful screen from over 500,000 compounds. Lastly, they compare the two methods in terms of reagents, equipment, cost, and issues.
