In Modern Hebrew, there is no practical difference, as the distinction has faded. In the pronunciation used here, the vowels with ע are pronounced in the back of the throat, which is likely the more historical pronunciation. The vowel sounds wjth א are pronounced more forward in the mouth, basically where we pronounce them in English. The cognate letter 'ayn ع in Arabic is still pronounced from the back of the throat if you want another example. One of the best ways I have heard to describe the sound is to imagine Arnold Schwarzenegger going "aaaahhhhh!" (If you're familiar with his yelling noises). Hope that's a helpful distinction.
