Sooo.. for DIC you still need phase contrast objectives (the ones with the ring inside the objective) or you can use a regular objective ?

This situation is incredibly frustrating. Numerous questions are being raised, but no satisfactory answers are being provided. It is vital for the medical community to address these questions openly and honestly if they wish to earn trust. While issuing a call to action is commendable, it is imperative to offer definitive solutions to these questions rather than simply admitting ignorance with statements like, "We don't know the answer to your question." What we truly need is a comprehensive approach that provides concrete answers. Moreover, physicians should be equipped to share the results of clinical trials with their patients, as withholding information and insisting on their own expertise is not an advisable method to obtain informed consent.

I personally lunched on a very helpful Serpa whilst trekking downward from Everest two years ago. A little gamey, yes, but I’d do it again. And, for what it’s worth, I sent his family a very generous tip in spite of the fact that he did not finish the trip.

Something doesn’t seem correct with that Bright Field demonstration 🤔 I’m fairly new to this, but if you’re using a 10x objective, then it seems like there should still have been SOMETHING that showed up in an unstained image (someone correct me if I’m wrong). It looked almost as if the image wasn’t adjusted for bright field during the condenser change , but if the condenser was changed, wouldn’t the lighting, camera exposure settings, and possibly even the focus need to be adjusted as well?

Excellent description of the two methods. It would be useful to compare images of the same specimen using the two methods to see what difference there are in resolving power for the respective methods. When do you use phase contrast in preference to DIC? Thanks

I'm not sure how touching the INSIDE of a sterile glove compromises it's sterility. That's why the cuffs are folded inside out. Now if she touched the OUTSIDE of the sterile glove with a bare hand, there would be an issue.

What do you use to clean your microscope? I just purchased an old Zeiss Axio Vert 100 and Objectives for my first ever microscope and love it 😊, but have found it quite difficult to get it clean and keep it clean. Also, tried cleaning the mirrors under the eyepieces and not sure if I did more harm or good... I put it all together from 2-3 KZfaq videos and no history using a microscope, with DIC and phase contrasting... I also purchased a Varel condenser/objective that I haven’t tried yet, and fluorescence DAPI/TRITC filters that I haven’t installed, which is when I’ll probably finally read the manual for safety reasons lol

LC-MS technology plays very crucial role in metabolomics and other biological fields. Conversely, metabolic profiling can give an instantaneous snapshot of the physiology of that cell. One of the challenges of systems biology and functional genomics is to integrate proteomic, transcriptomic, and metabolomic information to provide a better understanding of cellular biology.
