Wow!!! That is surprising results!!! PLEASE do the same thing with Lions Mane 🙏🏼😊 that would be extremely interesting to see. So many people are knocking myceliated grain spawn, saying that extract is 100% the way to go. 🤔

Next time send just extract of cordycepin not mixed with lions mane because the lions mane extract diluted the cordycepin for a more accurate testing.

Good for you and thank you for spending the money to do the testing. Mushroom vendors often provide misleading or confusing information. What you provide is straightforward and easy to follow. Indisputable facts are nice to have.

This is such a cool moment to be present for, as someone coming from a very amateurish ‘citizen scientist’ perspective it’s really interesting to see the arc of how this question arose and is now being addressed. Wonderful that a lab is up that can do the testing too, such a complex system that has to be erected to make something like this happen. A few thoughts: 1. Was the extract made from coryceps only or were there other mushrooms (in your video that is the case)? 2. For alcohol part of the extraction, you should use dried mushrooms (water in the mushrooms dilutes alcohol and decreases extraction), 3. You can evaporate some alcohol (and water too in the water part of the extraction) to get a more concentrated extract. Just my thoughts. I’m making extracts too and just got a chemist in to advise me on the best process. Boiling off and recovery of the alcohol with a vacuum could be helpful.

So 1. Watched the method he used to make the tincture, and it was just fresh fruit bodies if this is the same tincture as the video in the description. And I'm not surprised by the results, but the results were interesting and I really appreciate that he did this

Thanks for the video! Really appreciate you taking the time to create the content and the transparency of testing. A few notes which I'm sure you've figured out by now lol 1. Getting a test for a freeze-dried cordy only dual extract would have been a good first test to run for getting a more standard testing of cordycepin levels across the fruit bodies, cake, and extract 2. Either finely chopping, or putting them in a blender to break them down as much as possible will allow exponentially more surface area for contact for a better extraction 3. Like someone else said just to mention again, fruit bodies are largely water, so factor that into diluting your extract unless you use dried/freeze dried

Great video! Would love a cordyceps grain comparison video por favor (what grain cordyceps like best) and maybe even experiment with bugs mixed in with the grain (I’m doing such an experiment myself with meal worms) Btw I’ve read that cordyceps like alkaline conditions (9.0 ph apparently) which would help with any mold (mold likes acidic ofc)

Did you notice that you compared Paul's product which is not an extract but mycellium MIXED with Brown Rice, so not pure mycellium. You compared it to Oriveda's product which is not pure Fruiting Body but is Extracted (probably 6 to 1), unlike Paul Stamets product (not extracted), furthermore Oriveda's product is 2/3 MYCELIUM and 1/3 fruiting body so it's not even 100% fruiting body. Not an equal comparison of Fruiting Body vs Mycelium as you state in the beginning of your video. Would love to see this experiment between pure mycelium powder and pure fruiting body powder, (both extracted and not extracted), and without mycelium mixed in. Loved your video 🙏

Hushappy used black beans for his substrate, and had a 5x yield over brown rice, and his fruiting bodies 5x also..

reduce ethanol volume after extraction... use a rotovap to recover some ethanol and water and concentrate the targets.

That's excellent news! Now you know exactly where you need to put more effort and where you're doing well. I love that you're doing testing and sharing the results, right on. It'd be great to see the same kind of video done with the compounds in lions mane.

Also you might consider the amount of solvent your using for extractions. Also another thing to consider with all this testing, it's a matter of time before we figure out how many beneficial compounds are NOT water or alcohol soluble.

I have access at my job to utilize an HPLC, but I'm not sure where to start exactly with trying to test my own dual extracts. I'd love to but I need to be ready for testing before I ask my boss if I can have a day or two on the machine to test my own product. Do you have any recommendations on materials that might tell me about standards and methods? Love your videos and so happy to see how far your work has come. -Brittney

So cool. In your extract video, you used fresh and whole mushrooms, right? I'd imagine that the water weight of the mushroom is diluting the tincture from the start. Also, I think surface area would be an issue as well since the mushrooms are whole. The results for the mycelium really surprised me. I was thinking about how you'd incorporate that in a dual extract, because the water extraction process would just cook the rice and pollute the end-product. Since cordycepin is more alcohol soluble, maybe you could use mycelium for the alcohol extract, and fruit bodies for the water extract to get a broader range of things. Using a less porous and heartier substrate I think would be beneficial as well, like maybe corn kernels? The logic being that the alcohol won't penetrate as much and pollute the extract process. Maybe you could even tumble the mycelium off in a salad tosser, or just shaking a jar, then making an extract of that.

Your extraction process is probably all buggared up. Just soaking it isn't enough. Look into the temperature at which cordycepin degrades...if it's not below the boiling point of ethanol then maybe you can do a soxhlet extraction.

I understood it to be the grain after being dried out but not extracted in any way. It's got me thinking, if you're making your own tincture, that it'd be better to put both the myceliated grain and the fruiting body in your extraction solution

You mentioned a mix of reishi and lions mane in your double extraction. Could that factor in to the low cordycepin level. Would be interesting to see the side by side on extractions of just fruit bodies and mycelium both. Great video

Definitely could have. I'd love to see the results of a fruiting body compared to a cordyceps only tincture. And on top of that it'd be cool to see a test comparing different extraction times. Leaving the mushrooms in there to extract for several months seems like overkill to me

@garry have you considered ultrasonic cavitation for your concoction to break the chitin cellwall as a possibility? Lindsay Wilson has on youtube a neat, detailed explanation how to tune your own made probe and horn, if one has no ability to flex with highend labgrade or industrial unit. Are you going for vacuum destilation or spraydry the extraction in the end? regards0

As always need your valuable advice. Its been one month that my oyster mushroom is not pining all though the bag is completely colonize in my new fruiting room. I kept the temp in the fruiting room around 20'C to 22'C and humidity around 95 to 99. I also left the door open around 4-5 hrs everyday for fresh air. The cut i made in the bag swell like it going to fruit but it just turn brown without forming any mushroom. Want to know what am doing wrong.

try putting in the fridge for a few hours to shock it into fruiting it might not be a cool enough drop in temp. Also, maybe there is contamination from where you made the cut, try making a smaller hole with a clean blade on another area near the slice it may trigger pins

maybe it sould be considered calculating the dry weight of the cordyceps used to make the tincture and how much dry weight would correlate with the tested amount. maybe there is some compound lost/ left behind at the extraxtion process

I have a bunch of 1000ml Q45 media bottles that im going to use for LC.. should i have the lids solid tight on the jars when i pressure cook my honey solution ? or should i crack the seal a quarter turn or so and just cover it all with foil when i PC it all ? Still working on this issue - the latest method is dual extraction but it is very labor intensive. Some extraction companies claim to have bipolar diluents so Im going to try that next time I just need more fruits now 🫡

Really this is setting us up for a much broader understanding of our food in general. Like analyzing nutrients and minerals. Would be cool to compare a water extract to a alcohol extract through various mushrooms and see what's more water soluble and what's more alc soluble, and maybe even to test the remaining product after extraction. You can also evaporate some of the liquid with heat to get a more concentrated tincture to try increasing the concentration.

Amazing video Gary! Thats the way this industry must go. I would like to know how does the lab extract the cordicepin from the fruiting bodies and miceliated grain, since that could be a good extraction process for your tincture too. I started making mushroom tea and Teamster a couple years ago myself. I've always wished for a way to get my products tested both for potency or the presence of heavy metals. Because I Wild Harvest much of what I sell and consume. I've sent emails to the local universities and Laboratories but can't find anyone willing to work with me on this. hey QUESTION is peptone safe for human consumption if not what could I use instead for my cordyceps rice cakes in light of your results im gonna stop throwing them out and make them in to extracts and capsules but I can't find any info on lepton and human consumption

Just curious if a hot water extract alone would have given a higher cordycepin content. I know some mushrooms benefit more from singular extract via hot water right? I've read in a lot of places that Cordyceps are one of these. Perhaps the concept of mixing different shrooms together that require distinct forms of extractions negate the effects of the other tinctures mixed with it...? Would love to know

The water extracted sacharides fall out when you mix with alcohol. Reishi is the only common medicinal mushroom for which double extraction is worth it.

Could it be that you mix up Adenosine with ATP when you talk about metabolism? I mean yes, Adenosine is part of the biosynthesis pathway of ATP but primarily it's a nucleotide and therefore used in DNA synthesis.

No it's not at all "almost the same thing"! To compare the results you would need to know how much mushroom you used to produce your extraction and then convert all to grams used for extraction. Simply this result saiys nothing! For example if you took 1g Mushroom to produce 1ml extract you would need to multiply the extract result by 10 because percent is based on hundreds and mg (the exact result) is based on thousands. Also it should be clear that an extract can never have more concentration as the original product except when you use excessive amounts more mushrooms for extraction. For example: If you use 1g mushroom for extraction and get 1ml the 1ml extract will always have less content than the 1g of mushroom since you can never get all out. But if you use 1kg of mushrooms and conentate the extract down to also 1ml the extract probably will contain much more then the original mushrooms. I don't need a lab to know that its simple logic! So basically as soon as you do an extract and use the same amount of mushrooms it will ALWAYS be lower than the original mushroom if you don't concentrate it down later in the process!

yes “almost” is a relative term. It is a baseline for my process though and leaves lots of room for improvement. This is the first of many rounds of testing which will help me to dial in the process. As batches get better the weight of sample used etc. will increase so I wasn’t so concerned about these specifics.

This kinda is sad that the test for the tincture was done on your very first made weak tincture…The video is just starting to have value for tinctures as you can tell people there possible results from making the tincture like you did with a lot less mushroom to alcohol ratio compared to grinding up dried mushrooms getting a LOT more mushrooms in the mushroom to alcohol ratio..Honestly, I think that could be the weakest kind of tincture that can be made that you tested against the fruiting body..You might get the strongest by also evaporating or boiling of some of the solvents…

To be honest, this makes me wonder whether the other 2 compounds help with the absorption or effects of cordycepin. The active compound of a medicinal herb rarely achieves the same result as the whole herb itself. Not even talking about side effects when the active compound is on its own. However, the results of the dual extract was a big surprise.

Okay, but largely meaningless. That is 3 markers out of how many compounds? This reductionist “science” is severely flawed. It is comparable to looking at co2 as a measure of temperature. Need to look for the big picture and realize that the true nature is much more complex than we can imagine.

What if you would have snap freeze the Cordyceps: that is to use an small tube and put the tube in liquid nitrogen then take the tube and put it back to -80 freezer. That would have given you a higher concentration of your desired metabolite.

I give cordycep to my dad, he’s 68.. the results where absolutely life changing, he was doing really bad after almost passing from covid and other issues.. I gave him powdered fruiting bodies.. he wasn’t even mobile on a walker until he starting dosing cordyceps.
