It is for detecting, analyzing, and identifying proteins, similar to the western blot technique but differing in that protein samples are not separated electrophoretically but are spotted through circular templates directly onto the membrane. How do you get rid of the halos in the result, the one that you got in the end? I work in similar fashion everytime, but sometime I get good dots , but sometimes I end up with halos. What do you use for blocking?1% BSA when poured on a well without the antigen, yields signal when treated with primary and secondary antibody followed by the ECL mix that I use in my lab. What would you suggest to use for blocking other tahn BSA?

In the manual for the Bio-Dot apparatus it says halos are the result of the membrane not being properly rehydrated before applying samples OR excessive concentrations of sample being loaded

the result is not optimal. Also, the key control in turning on or off the vacuum was omitted. Good video, but can be more helpful for sure.
