Sir after saturation of liver the drug gets metabolized (900 mg)so it can also contribute to clearance additional to 100 mg ,altering maintenance dose..So sir what's the precision regarding maintenance dose.

Thanks dear. If drug is hydrophobic menas it is lipid soluble. So it can easily cross the membranes

Sir after saturation of liver , say after time 't ' when saturation decreses , the plasma concentration 100 mg is still remains or the plasma concentrations decreses after time 't' contributing saturation which has decreased.

The concentration of a drug goes on decreasing in plasma and the tissues dur to elimination How couldn't the maintenance dose does not depend upon the volume of distribution ( because no the organ will take the drug from plasma because of elimination of drug)

Hello, sir I have a question, cholorquine goes to liver is first pass effect , right ? Then why don't we avoid this way of giving dose? Why don't we use intravenous method of chloroquine to kill malaria pathogens ?

If for example 10 mg is given intrvenously ( 10 molecules of drug, each of 1mg).Now in bloodstream say:- --- 3mg is bound with plasma proteins --- 2mg/5L remains free. And --- remaining 5mg are distributed VoD =. 10mg - - - - - - 2mg/5L = 10mg. 5L/2mg = 25L Here we found that the drugs bound with plasma protein are not considered as they are also present in the body fluid even in the same fluid. Why they are not considered.. Plz aware me about..
