Hello dear Raja! Thanks for the tips on stain index! How about an antibody in which the marker expression causes a kind of smear or dim population then a bright positive population, how can we proceed in this case?

Hi there, thank you for the tutorial. Can you show us how you'd use the flowjo stain index calculator when running a titration of your conjugated antibody to determine the minimum Ab concentration with maximum signal?

Thanks Aja for sharing. Do you know why SDofNeg*2 is necessary in the Stain Index equation? Could we have simply compare the difference between the medians of + and - populations?

For the determining the resolution, you need to know the spread of the background. You could have huge differences in the medians of negative and positive- but if the negative has a huge amount of spread at that concentration as well, it’s not useful. That’s what the standard deviation accounts for and is a crucial part of the calculation

Thank you for sharing this information, however after doing stain index for each sample. Or running concatenation and have stain index of each concentration.How we suppose to pick the perfect concentration? Thank you

That's what the stain index is for! Whichever has the highest stain index is your sample with the best resolution.

one second, how u just get the SI in the excel? I don't think I follow that

That is a spreadsheet that I have made previously- it is simply where I put each of the required values for stain index calculation (MFI pos, MFI neg, robust SD negative) and another tile where I have the calculation setup to run
