Hello we have pcr thermocyler of analytic Jena biometra and in my pcr machine gradient pcr option is not showing anywhere I m very confused how can I set the gradient pcr?

I think 64.6 is the perfect annealing because 62.3 itself showing two bands mearged which is not in 64.6....!

Thank you. I still don't understand about duplex pcr. Duplex PCR has to be optimized for annealing temperature because it uses two types of primers at once. Can you explain about the duplex pcr process?
