The Lion's Mane project is trucking right along, and I finally have fully colonized grain spawn. I decided that I'd grow these mushrooms on an enriched hardwood fruiting block, made from soaked hardwood pellets and mixed with wheat bran and gypsum powder. After combining all the ingredients and loading them into the .5 micron filtered grow bag, I pressure sterilized the substrate for 2.5 hours at 15 PSIG. After allowing the substrate to cool to less than 85 F, I mixed in the fully colonized grain spawn.

Whatever you use should be dried up and not contain too many ingredients. Hardwood sawdust is the best. Some mushrooms like oyster thrive on wheat/paddy straw as well. I've heard that leaf litter also works for oyster mushrooms. Also 70% used coffee grounds mixed with 30% straw and even 50% Cocopeat mixed with 50% vermiculite (some even suggest adding gypsum to this) are other options.

Great video guide, thank you for this. One thing I would recommend as an alternative to grain-to-grain transfer is using lion's mane in liquid culture form - it can rapidly expand one's lion's mane culture and there is no risk of getting any substrate on the bag filter patch, it also sidesteps the necessity of a glove box or still air box and is generally quicker and easier when inoculating the bags.
@seancostello26
@seancostello26 3 жыл бұрын
you shouldnt use liquid culture on a fruiting block, that's only good for inoculating bags of rye grain, and then at some point you'll still have to do a transfer. He wasn't doing a grain to grain transfer here, he was transfering his fully colonized grain into a bag of substrate, which is something you'll have to do even if you start with a liquid culture.

Super helpful video, just got some fuel pellets and want to try this tek. The part where your camera died though, I'm hung up on that part of the process unfortunately :/ Do you pour the spawn on top and somehow move it around by pushing in different spots of the substrate bag? I'm just having trouble picturing it. I could obviously give it a shot and just free ball it, I know it can't be that complicated, but I guess what's the point of finding how-to videos on KZfaq if t you've got to do that anyways.

Hey, I just grew Lion's Mane this way and yes you just pour in the spawn and kind of squish it around and shake up the bag a lot:) It doesn't matter if it's not perfectly distributed because that seems hard to achieve - just do your best and the mycelium will take hold.

Thank you! I think the 5-6 pound bag is typically used because it strikes a balance between a few factors: -Sterilization time -Fit into the sterilizer -Colonization time -Ease of handling -For large scale growers, fit onto shelving in fruiting chamber. I think a larger bag would work but wouldn't be as efficient as the 5-6lb bag. But maybe I'll give it a shot and see how it works!

Hi. How important is moisture and air exchange during colonisation of the substrate?
@OneEarthMushrooms
@OneEarthMushrooms 3 жыл бұрын
Hi Laura! Having the correct moisture content in the substrate is pretty important; if it's too wet it could develop "wet rot", and if it's too dry, you may experience slow colonization or a failure to colonize at all. Air exchange isn't super important, you don't need any fresh air while colonizing. I use bags that have a filter patch to allow gasses (CO2 mainly) to slowly escape, preventing the concentration of CO2 from getting too high. CO2 is needed for colonization, but too much can be toxic to the mycelium. Thanks for watching!

90 minutes at 15 psi is sufficient for sterilization, and also, if you're using normal wood chips you have to sterilize, but if you're using hardwood pellets it's actually an unneccessary step as they are treated with a lot of heat and pressure during the forming process

Would the hardwood pellets not already be sterilised through the manufacturing process of turning the hardwood into pellets? I use straw pellets to grow various types of mushrooms and skip the pressure cooking of the substrate and have never had issues of contaminates. Do you think you absolutely need to pressure cook the substrate in the bag?
@OneEarthMushrooms
@OneEarthMushrooms 3 жыл бұрын
Bryan, honestly I don't think it's necessary. It doesn't make sense...the pellets are manufactured at a very high temperature and pressure, much higher than the thermal death point for thermophilic bacteria...pressure cooking seems unnecessary. I think the only thing that contributes to the potential for contamination in my process is the wheat bran, as I'm not sure what temperatures or conditions that material sees during processing. And yet, while I acknowledge all of this, I continue to pressure cook my bags. I'd like to take a more scientific approach to getting away from pressure cooking, and it's something I'll definitely be exploring more in the future. I haven't been very vocal about these goals, but I'd like to refine a process that doesn't use pressure cooking and doesn't use single-use plastics... but can still produce on a large enough scale for small time grower to be able to use it. Thanks for watching, and thank you for the thought provoking comment.
@bryankneis8286
@bryankneis8286 3 жыл бұрын
@@OneEarthMushrooms Thanks for the explanation on this. I personally haven't used wheat bran before but that makes sense that it is a source of potential contamination. If your thinking to skip the pressure cooking you could try and mix the grain spawn with the hardwood pellets and only use half the water. Then if you leave it for a couple of hours and add the rest of the water and wheat bran, you can provide the mycellium a head start in cultivating the substrate over competing bacteria. As long as your using something between 10 - 20% spawn to substrate ratio, this should minimize the chance of contamination. I live in the UK though, I think you mentioned living in hawaii so maybe you need to be more paranoid in that climate. Thanks for the great videos, just went through and watched some others on your channel. How do you use the lions mane mushroom? I hear they taste like sea food but have never tried it, only powered in a pill.
@OneEarthMushrooms
@OneEarthMushrooms 3 жыл бұрын
@@bryankneis8286 Hey, thats a pretty good idea! And another thing that sparked in my brain is to mix up the bran and half of the hardwood pellets and allow it to sit for a day...this should germinate most of the bacteria and fungi in the bran. Then on day 2, add boiling water and the remainder of the pellets. The boiling water should kill off the germinated organisms, then I can inoculate as soon as it's cool enough. I haven't eaten the lions mane whole yet. I dried and powdered all of the lions mane from this video, and put it in pills. If i get better quality fruits on my next bag, I'm going to try to make some seafood substitutes out of it.

Hole saw that is the size big enough for your arms to fit through or get a blue torch from lowes and a can of soup. After your meal Heat that bitch up a few times and press it and twist where you want the holes. Doesn't have to be red hot.

Your Gypsum calculation is based on wet weight. It should be based on dry weight. You're using 113g gypsum instead of 38g.

Can I cut a chunk out of my old grow block and put it in the new one for the mycilum to get new nutrition and fruit again
@FrozenFlames243
@FrozenFlames243 3 жыл бұрын
if its sterilize and there is no contams( usually have hidden contams). yes you can but it will not fruit rather it will go to colonization and once its fully colonize then it will go to fruit, best is either take a spore print or clone to agar and start from there.

Pine is a soft wood. I personally haven't used soft woods for lions mane, but they seem to grow only on hardwoods in nature.

I don't see why not. Is corn gluten free? I've used corn for grain spawn before; seems to work fine.

I hadn't considered contamination on the back of the patch (new to this). So I put a piece of micropore tape over the inside of the filter patch before filling the bag with the hydrated HWFP mix, and then used the chip clip idea mentioned below for mixing in the colonized grain.

Haha yeah a lot of people in the north use them for home heating! There are cheaper ones made from soft wood that work great for heating. The hardwoods (apple, maple, etc) are great for smoking.
