Hi there! HER2 has no identified ligand, it is always in an open conformation that allows dimerization with other receptors of the same family (HER1, HER3 and HER4) forming homo and heterodimers, and being HER2:HER3 and HER2:HER4 the heterodimers with most signaling activity. EGF is a ligand for EGFR/HER1.

You can add that HER-2 receptor is coded by the HER-2 gene which is a proto-oncogene , amplification of the HER-2 gene leads to a gain-of-function mutation.
