Bacteria can transfer DNA by means of three natural processes. They are transformation, transduction, and conjugation. Among three, conjugation is unique cause it requires direct contact between the donor bacterium and the recipient. Donor cells generally transfer F plasmid to the recipient. F plasmid can also be integrated into the chromosome and when integrated it can able to transfer chromosomal genes. Donor cell transfer chromosomal genes depending on the order of the genes and distance between the genes. This process helps us to map the bacterial genome. When we interrupt the mating process at a certain interval and examine the recipient for the gene which have entered at that time tells us the distance of that gene from the oriT.
Conjugation is studied in great detail in E. coli. In some national level examination, min mapping problems appeared frequently. So, I have solved some of them to clear the concept. I hope you will find this video helpful for your exam preparation. best of luck.

I couldn't understand how to put the arrow of hfr.

Imagine this is an arrow, so which portion hit the target first? Obviously the arrowhead. So the gene which entered first is situated close to arrowhead then second and so on. The last gene is situated at the tail. For example- ---a-b-c-> means c entered first then b and last one is a. But if ---c-b-a-> then a entered first then b and at last c. I hope now you can point the arrows in its proper direction.

Sir, could you please tell me how to consider orientation of the arrow (arrow head facing left or right). I did understand though how to arrange the genes on the arrow ie. first and last genes to be arranged starting from the head of the arrow till the tail of the arrow respectively.

It depends on the direction of F plasmid integration and OriT. For example X Y ----> ZA Means, Y 1st, X second and last is A then Z. If F plasmid integrated in this way XY is F plasmid

at 6:50 I did not understand drawing the arrow, based on what sir you draw Hfr 1 arrow going to the right but Hfr 2 going to the left?

Is there specific point where the plasmid will break before transfer. Or it can break at any point.

No, there is a specific point called oriT from this point F plasmid always breaks and start it's transfer. You can pause the slide where I have shown the map of F plasmid.

Are you serious or joking? PhD is a training and scholar work hard by himself/herself to establish a hypothesis. Where to do and which area to choose is possible. But, how to do PhD is impossible to describe. You will realize this after 1-2 years, when you start your PhD work.

No you are wrong. Look at the Hfr3 where e gene enters before g but after f. That means gene order is gefcdb--->
