"Delicately adjusting your vocal chords" and "putting all your oomph into screaming" are very much mutually exclusive. I've recognized actors of remarkably different characters just because of their screams; Robin Atkin Downes can't help but use the exact same death shrieks for a German doctor and a Japanese-American soldier.

Its not really just "loud = funny" but "unexpected but accurate = funny". You don't expect the characters to just turn into raid sirens, but they usually do it to convey a serious emotion, in which the exaggeration helps convey it.

Fun fact: the reason the whole "if you're late one more time you're fired" line was used twice was because the shorts shared the exact same plot; WB completely remade "Porky's Badtime Story" into a color version called "Tick Tock Tuckered", replacing Gabby Goat with Daffy Duck (which automatically makes it the superior short)

Blanc reprised his roles of Bugs, Daffy, Porky, Sylvester and Tweety for 1988’s Who Frames Roger Rabbit, but famously could no longer voice Yosemite Sam because his screams were too difficult for the aging legend to recreate. With screams like these, it’s a wonder his voice lasted as long as it did!

Mel Blanc's loud screams is a textbook example of how to do "loud = humor" comedy right, and he is what modern comedians should be. But to be honest, he didn't had to go hard on his loud screaming, but he did. And we all should be thanking him for that.

The "SMOG!" at 5:26 is because Arthur Q. Bryan (Elmer) couldn't do a powerful enough version, so Mel did it instead.

The phrase came from the "lights out" requirements that were held in towns around the country during the war. The authorities wanted everyone's lights out (or their windows covered with black curtains) so that if any enemy planes got into US airspace, the wouldn't know there was a town to bomb below.
