Metabolomic workflows mixing high resolution OrbitrapTMplatforms and low resolution triple quadrupole platforms with data analysis from targeted, untargeted and mixed mode methods will be demonstrated and explained using plant metabolomic data that has been created here at the University of Florida through collaborations with Thermo Fisher Scientific. Modern Vanquish™ Horizon UHPLC Systems employing reverse phase, HILIC, and hydrophobic long chain stationary phases will show how multiple chromatographic approaches can increase the depth of identified endogenous metabolites with high precision quantification. Library building with TraceFinderTM and XcaliburTM will complement untargeted analysis using Compound DiscovererTM, Lipid SearchTM, and mzCloudTM and will be used to drive mixed mode data dependent methods utilizing ion trapping type instruments.
