What is a Differential Amplifier:
A differential amplifier amplifies the difference between the two input signals. It is widely used in integrated circuits as an amplifier.
The first stage of the op-amp is also a differential amplifier.
In a MOS differential pair, two perfectly matched MOSFETs with identical characteristics are used.
In the integrated circuits, it is possible to match the characteristics of the two MOSFETs to some extent.
Advantages of Differential Amplifier:
1) The ideal differential amplifier eliminates the common-mode signal at the output and only amplifies the differential input. Using the differential amplifier it is possible to eliminate or reduce any noise or interference which is common to both input terminals.
2) When multiple differential amplifier stages are cascaded together, then it also eliminates the use of coupling capacitors between each stage.

what is the advantage of common mode signal input then , if the differential output is 0 all the time. where it is used ?

You need to have a DC Signal (Common Mode Signal) in order for current to flow. The common mode signal essentially turns on both FETs and turns them both on the same degree. Adding small AC input signals on top of that will ensure that there is a differential output at the drains of the transistors.

How is it that Vo1 and Vo2 have different polarity when it connected the exact same way?

The polarity of Vo1 and Vo2 is different for only differential input. For the common mode input, it is same. And that's why for the common mode input, the differential output Vo1 - Vo2 is zero.

If true differential input signal is not readily available, then the differential input signal can be applied at only side. And still, it is possible to obtain the similar result. For more info check this simulation. www.multisim.com/content/WfWiSqnoRcTvjcpFrX3KL9/mos-differential-amplifier/ Here, DC voltage on both side acts like a common mode input to both terminals. While, 10 mV input signal is applied at only one input. The Differential gain is 4. So, differential output Vo1 - Vo2 is 40 mV.

Yes, with the help of a transformer, it is possible to generate the differential signal from a single ended signal.
