If I move the plano convex lens upward then what should be the spacing between the plano convex lens and glass plate to observe centre ring as bright ring ?

Spacing should be multiple of wavelength/2.

Why don't we measure wavelength using only one diameter that is lambda = (square of Nth ring Diameter) divided by (4 N R)

Yes we can. But we take more readings to eliminate measurement errors.
