So now that you've done that your going to want to stock up on Saisetsu-shin's Sake for increased amrita gain, on Way of the Nioh with the Sake you will earn 16,860,745,722 per run but don't worry if you don't have the sake you'll still earn 7,202,398,066 amrita per run. To reach max level (750) Your going to need 1,599,152,427,363 amrita. If your going as fast as me and with load times you should be able to do 40 runs per hour. With a large stock of sake your looking at 95 runs or 2 hours and 22 minutes, without the sake your looking at 222 runs or roughly 5.5 hours. Time will vary depending on difficulty.
