Cre-loxP system is a highly efficient system to create transgenic mice. It relies on the ability of Cre recombinase to bind and recombine sequences flanked by loxP sites. Using Cre-loxP system, it is possible to study gene function by either activating or inactivating a gene in a tissue-specific manner. The gene can also be inactivated if an inducible promoter is used, e.g. Doxycycline or Tetracycline-based promoters. The system has been used to create mouse models of several human diseases and has led to a better molecular understanding of many diseases.

A doubt: Here the Alb-Cre is in a hemizygous state and also in a different chromosome?

Gayatree, the Alb-Cre can be in a hemizygous or homozygous state depending upon researcher's experimental setup, and yes, it is on a different chromosome. Since a variety of these mice are available carrying Cre-lox construct on different chromosomes, knockouts can be easily generated for genes present on different chromosomes.

Since hemizygous mice have a wildtype chromosome present, using hemizygous instead of homozygous Cre genotypes might minimize unintended consequences of random transgene insertion. (Courtesy: From Jax site)

Isn't there a way to design a Cre-Lox system that copies itself onto the homologous chromosome so that every offspring between a transgenic individual and a wild-type individual will be a transgenic individual? A "gene drive"?
