As far as I understood from your video that in case of bacteria when there is a tetracycline present in bac cell tet resistant gene is expressed then why in case tet-dependent gene expression is not seen in case presence pf tetracyclin?

why cross two mouse with rtTA/TRE respectively to get tet-on system? Isn't it easier to acquire a tet system by transforming rtTA and TRE into Embyonic Stem cell using CRISPR?

Very helpfull video :) Can a gene be expressed in a tet-on system if no tetracyclines are present? Or will there be absolutaly no expression of the gene if there are no tetracyclines?
