That's a great question, Amanda! If the male in question is still living, I suggest he begin with an atDNA test from Ancestry. Ancestry has the largest matching DNA database of all the companies. Once the results are in, he might be lucky enough to find a close relative like a sibling, parent, or grandparent. If not, cousin matches along with traditional genealogy research may flesh out his biological parents. Ahh...a very good question and not an easy answer. If your Native American ancestor was very recent, it may show up on any of the atDNA tests. However, as I explained in the video, ethnicities are estimates based on the reference population of the given company and how much of that particular DNA you inherited from your parent(s). When dealing with proving Native American ancestors, it is best to do old fashion genealogy research.
