We can use a sodium bicarbonate(Nahso3) because it extracts carbon dioxide.

No, you need to break the upper epidermis layer of the leaf as well as it's chlorophyll pigment.

But is it okay if you just boil the leaf and then test in iodine after de starch for 48 hours or do you have to do seal it with KOH, if I just boil it and test in iodine for will it still prove anything?

Why did you boiled the leaf

Because of when we heat the leaf the chlorophyll will get removed so we can see that the starch is present or not(when we not heat the leaf we cant able to see that the starch is present or not)after that when we drop the iodine solution with help of dropper it will change the colour. To see this we should heat the leaf with methillated spirit.
