In this video I am making one of the coldest/iciest/mintiest substances known, Icilin. It is a super-agonist of the TRMP8 ion-channel which is a receptor on our mucous membranes that gives us the cold feeling from temperature and food such as mint. The compound is not found in nature and is fully synthetic, there are no other known natural compounds that induce a colder feeling. Supposedly the compound WS-12 should feel colder but is less potent, which I will make later to compare.

I wonder if the high cost of certain chemicals (like Icilin) is due to lack of economy of scale. That is, you're essentially paying some other chemist to make your specific order just for you, rather than paying some amount from a much larger batch (possibly made by the same chemist).

Very likely. There is not really a market for it so vendors can also get away with charging ridiculous prices.

To some degree, yes. But also, it's a useless chemical to a chemist, who could easily make it themselves anyway. So the only ones who would be interested in the molecule are molecular biologists, and those people generally don't need more than a few mg to do an experiment, so suppliers are more than happy to charge the same price for 10 mg that one would for 10 g if they were selling to a chemist.

Random but I really hope someday a talented chemist will be able to discover a pain killer that DOESN'T cause negative physical reactions once someone stops using it. If someone can discover that it could genuinely change the world and improve so many peoples lifes. People that have never experienced physical withdrawals don't fully understand. Someone can fully be mentally ready but no matter what you do, physical withdrawals are completely unavoidable and it makes you feel like an utter failure and our society has a very misunderstood negative perspective on physical withdrawals and physical dependency. I truly believe if some scientist could discover something that doesn't cause physical dependency but helps with pain relief, then I really think it could positively impact the world.

I'm surprised you experienced such a mild taste from the DMSO. I got a tiny drop of it (lab grade) on my skin and a minute later there was a disgusting onion/garlic taste in my mouth. I also visited a veterinary barn where one of the equine patients with septicemia/toxic shock had been given 20cc IV DMSO and the whole place stank to high heaven for the rest of the day.

DMSO itself shouldn't have much of a direct taste when pure, it just contains sulfide impurities that make it taste a bit like dirty vegetables. I didn't have an aftereffect of garlic taste, I think that could be genetic

@@Chemiolis This was DMSO from Sigma-Aldrich which they claim is ≥99.9%. Yeah, it could be genetic as you say. I remember someone telling me that sensitivity to HCN is genetic. I can always tell immediately if someone in the lab has been using it, even when they can't smell it themselves. Mind you, the smell receptors also quickly desensitize to (or are killed off by?!) cyanide.

Apparently "Part of the DMSO is transformed to the volatile metabolite dimethyl sulfide, which gives a characteristic garlic- or oyster-like smell when excreted through the lungs", according to Adverse reactions of dimethyl sulfoxide in humans: a systematic review (10.12688/f1000research.16642.2).

I've got an exercise in awful for you to try. Isolate whatever is in a green persimmon, the small variety, that instantly dries the mouth and even the teeth, and taste that. I've never experienced something else that could make my teeth dry.

First time I tried a persimmon I thought I was having an allergic reaction from how bitter it was and how it made my mouth dry. I only found out after googling that the it was because I ate a unripe one. I still haven't tried to eat a ripe one out of fear of getting that bitter taste again.

Very cool heterocyclic chemistry! More of that please! For the dihydropyrimidinone reaction, maybe you could have avoided the initial precipitation and vigorous HOCN gas evolution (apparently poisonous) by adding KOCN into the AcOH solution of your starting material which would allow for a controlled formation of HOCN. Then add HCl to drive the elimination of water. But again, love the target molecules you're making. Keep it up!

The initial precipitation was not part of the plan! I would have just added it in portions into stirring liquid, but no. Either way it was fine, this reaction is very fast.

Erythritol is similar, I think. That slight “coolness” is gives to chocolate makes me think it activates the same receptors.

Please synthesise conolidine! No lab will make it, it's too complicated they say. It's not illegal, it's a calcium channel modulator and ack3 blocker, it's a fascinating molecule. Con-ol-idine. It comes from Tabernaemontana Divaricata plants. There is a published write up for its synthesis. It uses unusual metal salts in the synthesis.

Good old DMSO - always enjoyed how quickly the taste of oysters appears when it touches skin. Usually advised to use 85% aqueous since it's hard on your skin if it's pure....(some people use it as a topical analgesic, but the readily available MSM (methylsulfonylmethane) apparently does the same thing....did you know that MSM was briefly a controlled substance in California?

A little suggestion: using your Eppendorf pipette for highly corrosive and volatile liquids like conc. HCl will dramatically decrease its lifetime because it can corrode the mechanical parts inside. At least that's what's been told during my undergrad.

Have you tried making candies with it? Like heating it up with a little bit of water and a lot of Saccharose (and maybe some glucose) till all the sugar melts Then letting cool down in small pieces (basically making menthol candy on steroids) The this could distribute it rather well in the sugar and make it easier for the rezeptors to detect

Could it be that the reaction of acetophenol is driven by anchimeric assistance (aka neighbouring group participation) of the carbonyl oxygen? This would form an oxocarbenium intermediate, which is very sensitive towards nucleophilic substitution and would definitely allow this specific substitution to occur at mild conditions.

i wonder if 'chewing gum' companies like extra/wrigleys/orbit etc would love this stuff to be encapulated in some kind of nanoparticle to make a chewing gum that would last forever or at least much longer than anything out there right now.the cold peppermind and wintergreen sensation is what everyone loves and associates with the flavor lasting a long time. so interesting!

I draw them myself and usually let someone else check them. If I don't know it I ask someone. Often it is not readily found online and we draw the most reasonable/likely explanation.

Okay but how do you predict the way molecules will rearrange themselves within the right conditions with the right reagents? How do you ensure the chemical bonds as intended? Is it simply chance, followed by a gleaning of the intended chemical?

Heat? Very interesting. I have some menthyl methyl lactate, I wonder how this compares. I'm guessing the heat is because both propylene glycol and DMSO are causing a heating sensation when they pull moisture from your mouth, or are actually heating up from exotherm.

I was stoked to see this and immediately wondered "what about the WS- compounds?" Interesting historical note, the WS in WS-5, WS-12, and WS-23 alludes to "Wilkinson Sword", a shaving products brand that evolved from a gun manufacturer in Great Britain started around 1775. Apparently they wanted better cooling sensation compounds for things like shaving creams and discovered some that are now in chewing gum and pretty much everything else. Which brings me to the question: Why doesn't Japan approve amides as food additives?

I think the problem with a lot of amides is that they can enzymatically hydrolyse in the body to give back the amine that is usually toxic.

DMSO with WS 12 may have potential as a pain killer. However the enhanced solubility of DMSO may lead to toxicity. I don't know if it been greatly researched? I plan to show how to make DMSO, copper nanoparticles and spearmint essential oil extract.

WS-12 is used in vapes. This might be able to be used too, but with a nitro group being present it might not be very heat stable? Just at a glance, WS-12 looks like a more stable molecule that won’t polymerise or decompose readily under heating, unlike this Icilin

Well that was a good peek into the world of chemistry, My probably (at least to the best of my own perception, grades aside) least understood school subject, that I've never further build upon after end of High School. Considering how most of what is going on inside humans is affected by exactly stuffs like that... Imagine what the people that understand, understand this science do... and can do... Like that was a week long process by a single person to make an bubble gum compound on steroids, now multiply that by 100ths of staff members of well funded government orgs operated for years upon years, having all the previous research and know how available. Well ~or private.

WS-23 is one I hate, its frequently in E-Liquids and the cold sensation is waayyyy to strong in the throat it actually hurts breathing in after if you vape a good amount

is it safe to randomly taste a chemical without doing some tests before? i dont know anyzhing about chemistry but to me it seems unsafe/dangerous. wasnt one of the chemical something with cyanide?

No, you have to analyze it or know for sure that it is fine. In this case I did a simple but effective test and found it only contains the desired product. It is also important to take into account tasting vs ingesting, I always spit it out afterward. Besides, the amount is extremely small, even if my product was 95% pure, it is unlikely to cause harm. There aren't many compounds that are toxic enough to case harm with several milligram exposure. Experience and understanding the reaction and product is also ideal. With that, you can tell that there is no reason to suspect anything dangerous. There are more factors to take into account, such as literature precedent (the discovers tasted and ingested it too) etc.

Did you characterize it? Maybe the yield is so high because of some unsoluble impurities

Clean with only one spot on TLCs with Cerium phosphomolybdic acid stain. I don’t see a reason to expect impurities here either. Precursors wash out earlier and Icilin is cleaned easily in the end with its poor solubility.

​@@Chemiolis idk, just strange that a "add everything and mix step" resulted in a x2 yield compared to literature. Maybe they didn't break the solid precipitated mass of the last passage, so that mixing was suboptimal

I would’ve done a column before tasting it just to be sure 😂 Also, is it legal to sell chemicals for ingestion? Is there a license for that? Do you have to use food grade precursors? Or is it okay to as long as you don’t call it a food/flavouring agent/medicine? I’m genuinely curious, because I have an interest in the niche flavour compound market. Thanks

To sell stuff marked for human consumption you need certified lab, certified precursors and to have the entire process reviewed,and then have analysis done on each product batch - that's in EU, and I assume the US has equally if not more stringent regulations. Which is why nobody bothers, same with cosmetic ingredients.

Chemistry is very interesting, but in indian high schools we have to just remember organic name reaction and whole s,p,d,f block elements reaction. We learn 99.5% Chemistry theoretically. Teacher asks Chemistry practical question svia theory 😔. No one gives importance to practical and experiment...

Paraformaldehyde is less toxic, doesn't stink as much and it's a solid at room temperature while formaldehyde is a gas. So it's easier and safer to handle.
