In this video, we review the basic concepts of OTU and ASV-based methods of targeted microbiome sequencing resolution. Topics will include a basic introduction to targeted microbiome sequencing, the methods and logic behind both OTU and ASV generation, and the advantages and disadvantages of each method for 16S and other targeted sequencing studies.

Thanks, thats a brilliant video. The last point made me smile. How many times have we all read "OTU grouping" autocorrected to "OUT grouping".

how do we know if the sequence is or isn’t conserved?

If i were to use ASV, do I still need to run DADA2 or amplicon denoise before hand?

When you run your sequences using DADA2 (I used the DADA2 plugin in QIIME2), your output are ASVs.
