I noticed you always pause to interrupt that lady with the pitchfork reight before the Gigante boss fight. I'm fairly certain you can just run right past her if you stick to the left side and still grab the herb. I remember dodging her when I was doing my casual Professional run. Might save you like half a second or something which to a casual player doesn't mean much but to a speedrunner it might. Edit: Congrats on the sub 45, that's insane.
