Structural Activity Relationship (SAR) of Barbiturates

Activity and duration of action of barbiturates is highly related to the groups at 5th position in barbiturates. Interestingly the total number of carbons, nature of chain, branching and presence of unsaturation all these factors at 5th position greatly influence lipophilicity of the drug thereby affect the duration.

Barbiturates are an awesome example of what the power of simple carbon chains can do for a molecule , the ring / ketones are obviously the functional parts but without those elaborate carbon chains the molecule is useless

N alkylation increase anticonvulsant activity and large alkyl have sedative hypnotic effect and only one of N can be substituted
