Homelander is only a human who had his DNA altered by drugs. Superman is a genetically superior alien who is powered by sunlight. Homelander couldn't even save a plane load of people while Superman routinely makes such super saves!

Homelander would use Kryptonite! Fast speed and skills of delivery, even in Superman's body cavities! Sharpened pieces by Homie's powers! Before he was finished, Lois, Jimmy, and his parents may also have been krispy crittered as well! Just the numbers of a campaign where Homelander would be well informed about powers and weaknesses of Superman! If he doesn't play, he would win! Otherwise, I think Superman is many more levels ahead!

Superman was in fact trained in martial arts by batman and wonder woman and learned kryptonian martial marts, so yeah he's very proficient in hand to hand. Superman's love for Lois is actually one of his greatest strengths and has been the driving force for some of his greatest feats. Kryptonite wouldn't really help homelander either, superman once fought a doomsday while doomsday had evolved to have kryptonite bones. If superman needs to he can power through kryptonite, even become immune to it if he absorbs enough solar radiation. Superman's mental blocks and desire to not hurt anyone cause him to hold back a lot, though there is a comic where he tries to get past this. Zack Snyder's movies actually did the best in showing how powerful superman is and got him the closest to his comic book strength.
