@JP No, GCI detects signals (based on a phase shift) whenever the refractive index (RI) close to the sensor surface changes. This can be due to a refractive index solution bulk (RI difference of injected solution vs the running buffer), molecules adsorbing or binding (ligand capturing, analyte binding to the ligand) or even due to rearrangements or dynamics of the ligand or the sensor matrix. Any binding assay for characterizing a specific interaction is dependent on correct double-referencing of the raw signals (injection onto a reference channel without the specific ligand & blank injections) to determine the specific binding response. These concepts are very similar to how SPR technologies are used.
