Endocannabinoids and Mental Health
#CBD #Mentalhealth #cannabis
Dr. Dawn-Elise Snipes
Objectives
~ Explore the role of the endocannabinoid system in
~ Mood disorders
~ Cognitive disorders
~ Autoimmune and inflammatory disorders
~ Pain
~ Stress
Intro
~ The eCB system targets both monoamines and the HPA axis, which is also dysregulated during stress-induced psychiatric disorders
~ Hyperactivity of endocannabinoid signaling contributes to excessive intake and storage of high-calorie foods, while endocannabinoid hypoactivity is a risk factor for the development of depression, anxiety and post-traumatic stress disorder
~ The ECS is comprised of endogenous cannabinoids (endocannabinoids), cannabinoid receptors, and the enzymes responsible for the synthesis and degradation of endocannabinoids
~ Endogenous cannabinoids are endogenous lipids that engage cannabinoid receptors
~ The first discovered and best-characterized endocannabinoids are anandamide (arachidonoyl ethanolamide) and 2-arachidonoyl glycerol (2-AG)
~ Both 2 AG and anandamide are agonists for CB1 and CB2 receptors
~ In systems with low receptor expression, anandamide can antagonize the effects of more efficacious agonists
~ On demand, endocannabinoids are liberated in and released into the extracellular space. This contrasts with classical neurotransmitters that are synthesized ahead of time and stored in synaptic vesicles
~ CB1 receptors are abundant in the central nervous system (CNS), particularly in cortex, basal ganglia, hippocampus, and cerebellum
~ CB2 receptors are expressed at much lower levels in the CNS
~ Physical exercise mobilizes endocannabinoids, which could contribute to refilling of energy stores and also to the analgesic and mood-elevating effects of exercise.
~ A second important role for endocannabinoid signaling is to restore homeostasis following stress
ECS and Neurotransmitters
~ Changes in the ECS are involved in several psychiatric disorders.
~ Serotonin
~ eCBs reduce serotonin release in the CNS via the activation of CB1 receptors
~ Early evidence for a role of eCB signaling in the regulation of the 5-HT system suggests a high level of functional overlap between the 5-HT and eCB systems. Both 5-HT and eCB systems regulate body temperature, feeding, sleep and arousal and emotional processes
~ Dopamine
~ CB1 receptors tightly regulate DA neuronal activity
~ CB1 receptors are expressed at presynaptic inputs to DA neurons and facilitate or suppress DAergic neuron activity depending on their location
Alzheimer’s and Neurodegenerative Disorders
~ Cannabinoid receptor agonists such as AEA have been shown to provide protection against neurotoxicity
~ Activation of CB1 and CB2 attenuates Aβ (beta amyloid)-induced neuroinflammation, neurodegeneration and spatial memory impairment
Pain and Inflammation
~ Activation of the CB2 and adenosine receptors leads to a decrease oxidative stress and inflammation
~ CBD is one of the main pharmacologically active phytocannabinoids
~ It is non-psychoactive, but has many beneficial pharmacological effects, including anti-inflammatory, antioxidant, anxiolytic, antidepressant, antipsychotic, and anticonvulsant properties
Anxiety
~ Chronic stress and steroid treatments are both known to impair endocannabinoid signaling at multiple levels,
~ Stress-related down-regulation of the CB1 receptor leads to greater changes in glutamatergic signaling and excitotoxicity within the PFC following chronic stress
~ It has been repeatedly shown that CBD and THC concurrently decrease anxiety and depression
~ Essential oils, terpenes can activate CB1 and CB2 receptors and are associated with cognitive and mood effects.
Interventions
~ Omega-3 and 6 fatty acids
~ Flavonoids which inhibit endocannabinoid degradation
~ Meditation
~ Exercise
~ Sex
Summary
~ The endocannabinoid system interacts with most other bodily functions and neurotransmitters.
~ Problems in the ECS are associated with
~ Chronic pain and inflammation
~ GI disorders
~ Mood disorders
~ Neurodegenerative disorders
~ Metabolic disorders
~ Endocannabinoids are created partially from lipids, particularly Omega 3 and 6s
~ Reducing HPA-Axis activation and increasing serotonin and dopamine will all increase EC levels

Day 1 without marijuana. I've smoked about 2g per day going on 10 years. My stomach is a total mess this morning. I could only sleep for about 4 hours. I have a strong craving and my brain won't stop suggesting a quick call to my dealer. Chances are very slim that I'll make it. I ask myself why I would put myself through this for such a small chance of success... but being dependent on marijuana is so much worse than any of that. I'll take this small chance and if I miss I'll just roll the dice again until I hit. I won't allow another decade to be altered by my addiction. I'm ready to find a wife and I'm building my retirement. Those are not things to do while you're stoned. I don't want to spend the rest of my life wondering who I might have wound up with if I was sober... or what better career decisions I might have made. I don't need to feel good all the time, I need to be effective. I have important work to do.

Around 37:50 your commercial is starting when you are speaking. I love this so far, thanks so much. I really really like this it gives me hope. I have horrible food allergies and gut issues constantly. I also poo blood almost every time. They say it is hemmeroids but seriously it has been a long time. I have ADHypoD and the more I learn about bpd I might have been suffering with it as well. My daughter/son has BPD traits and is 16, so I have been obsessing about trying to learn about it, and how to deal with it. She is a quiet bpd. I used to smoke a lot of weed in the late 90s and 2000s. So much guilt and hearing about these kinds of things help me feel as though I was self medicating, and that shouldn't have been frowned upon. Though it has messed with my asthma. I might look into getting a script for something though I might need to travel an 1.50 hours away because in a place like this, everything is still pretty backward, even though it has been legal in Ohio for quite some time. Thanks

Guys, HIGH doses of THC WILL (not can), WILL reduce GABA synaptic transmission as well as glutamate within the brain. So if your focus is anti-anxiety, stop lying to yourself that high doses of THC help. It doesn’t for anxiety. It lowers anandamide also. Great advice on this video. Use a 1:1 ratio or at least add some CBD with your joints or preparations. With CBD you’re getting your GABAergic allosteric modulation, antidepressant effects due to the 5ht binding and the modulation of immune cells like microglia. Great vid!!

THC seems (keyword, seems) to be a double edged sword when it comes to anxiety, in low dose studies there is an indication of reduced anxiety, presumably through predominant glutamate and other excitatory inhibition (just my theory), rather than predominant GABA inhibition at that dose. However when high THC doses are studied, it seems that GABA is inhibited enough to cause an anxiogenic effect instead of anxiolytic effect and can induce anxiety attacks, so yes i agree here. CBD is extremely important and unfortunately most of the products out there only have predominantly THC with very little CBD.

Most in here probably already know this, but exogenous cannabinoids out comped endogenous cannabinoids at anandimide receptors by a thousand fold, so If you want better emotional regulation in the long run, then sober is the way to go. I've been a heavy user for two and a half years and stopped a week ago wish me luck.

@@DocSnipes thanks, i found the portion on endocannabinoids increasing serotonin which boosts dopamine, usefull specifically how neurochemicals influence eachother it isn't just about raising one neurotransmitter in isolation. Also the sleep part and part on gut microorganisms producing the bulk of you serotonin, I probably underestimate how much diet could improve my mood.

Hi. I am a 42 year old Irish guy, and have suffered from birth with my body trying and not being able to destroy the virus in Chronic sinusitis, thus leading to FMS/Depression. This video is the best I have seen in 25 years. I recently recovered from the COVID virus, my FMS symptoms and firing glutamic acid CNS multiplied by about 5.Therefore my conclusion is that the autoimmune hypothesis on FMs is true. Thankyou for this video and sharing of this information.

I learned from and enjoyed the perspective of psychiatric disorder treatments via the use of cannabinoids. However, I am skeptical about the medicalization of issues which I believe are fundamentally social and economic in creation, i.e. moral and situational rather than biological in origin. But I am willing to be convinced otherwise and 'there is more than one way to skin a cat'.

Hi, Doc Snipes, are you a medical doctor? Corticosteroids depress the HPA axis and not upregulate it and that is why it has to be tapered off to allow the HPA axis time to recover. Please research and correct it. Rest of of presentation is fine but this fact makes the whole presentation sound dubious. Please reply as just trying to help.

Wow, lots of great info. Please contact me, share some neuro cannabinoid science . If one tries to straddle the fence rail... eg even CONSIDER pHarma is ok... denigrates the total control the eCN system has over all systems. Perhaps a small distinction, but let's set things right and never have to use a rear view. Psychiatry has been since 1858 Eugenics. Benzodiazepines... Good grief . Hitler was bad enough with fluoride.. you say ok fluoride AND life addicting benzodiazepines are ok and that cannabinoids can fix this too? Straddle the fence and ya lose ability to walk.choose natural remedy and see how that heals. Try it then yammer
