Please answer>>>>I am a ex-soldier and have had some small CTE and I read some huge study that was not in America and it stated NMN worked on Dementia patients and even found some synapses/brain repair at a cellular level...I am not sure but I think I read 2 for mice and 1 for human with mild dementia, so that is why I have been taking NMN since they say it is a smaller chemical compound that creates NAD+ and I have seen results with short term memory. I also found out by my seller that the FDA is wanting to make a prescription out of it...guess who was named backing that, Dr. Sinclair. Can you explain this. Thanks and God Bless

I think we need research replicating Michael Lustgarten niacin research. It suggests that 60mg of niacin is about as effective as 1g of nmn, but less than that effectiveness drops at least for him personally. It may be that there's a minimum dose based on body size for effectiveness.

Thank you, Dr.Stanfield. I’ve been very critical of some of your past observations/conclusions that seemed to simply parrot orthodox views. In this instance, I appreciate the time you’ve taken to critically review these studies. It brings to the top of mind - for me - that as well trained as I am in statistics & experimental methods, I still easily revert to unconsciously accepting statements by “experts,” and anecdotal reviews. Not doing so requires constant reminders we need to continually wake up to going on automatic pilot. In all areas of our lives. And to do that, we must CONSTANTLY expose even our most deeply held thoughts, judgments or conclusions to rigorous and fearless scrutiny and challenge. Again…and again. And one more thing: it reminds me that just because I don’t agree with some ideas from another person, doesn’t mean anything beyond that specific thing. It should not be concluded that I am “right” such that I cannot learn and grow from that person (or most anyone else). And also, maybe I’m not even right. The more we can find joy in this journey-of curiosity, discovery, including being wrong, and learning and growing, the better everything is. I value your time and work. Thank you. thank you for these reviews. I used to be very enthusiastic about supplement recommendations, hoping for miraculous results. a thorough breakdown of research with emphasis on following the guidelines instead of the hype is very sobering for me.

I had complete visual failure.. eyesight became kaleidoscope, would last for 10 seconds.. eventually realized it was the Astaxanthin ... never an issue since stopping, except once when I didn't know it was in my omega sup.

He did an episode on Astaxanthin. My recollection was that there was good evidence of benefits, however taking excessive antioxidants might have downsides so he recommended it for those 60 and over.

First of all - THANK YOU FOR RETIRING THE WHITE SHIRT 😀 About your video - I tried NMN for 6 months, didn't really "feel" anything. Did more reading with Niagen and the work Dr. Charles Brenner has done and did that for 18 months, 300mg a day and then on up to their latest 1000mg offering. I've spent a good chunk of money and I just don't see it doing much. I see a lot of fans of each claiming they look and feel younger. I get that result from daily exercise, collagen, hyaluronic acid, creatine and other supplements you suggest. So despite some fascinating reading, there isn't a magic pill to turn back or slow down the aging clock quite yet! (P.S. I can't wait to see Marcel's rebuttal lol)

Glucosamine sulfate is effective, according to several meta analyses. Studies also including glucosamine hci fail to find an effect. Details matter. Combining collagen hydrolysate and glucosamine sulfate is probably a very good idea to prevent cartilage degeneration. One should also not confuse cartilage regeneration and joint pain, because joint pain can have different causes, such as muscular or connective disbalances, or inflammation at other parts of the joint aside of cartilage. Maybe damaged cartilage started the inflammation cycle, but then other tissues started to get inflamed as well and continue to be after cartilage issues have been resolved, and same goes for inflammation because of muscular and structural imbalances which could be caused by pain from cartilage damage in the first place but stay even if cartilage has been regenerated. So to resolve joint pain, just improving cartilage often is not enough and is not a reliable marker for if a supplement improves cartilage or not. Again, details matter. Tip for anyone with joint inflammation, try taurine, it really helped me. Still supporting my cartilage with collagen hydrolysate and glucosamine sulfate though. Also, glucosamine sulfate is so cheap, cost is not an issue as well.

There’s no biological pathway for orally supplemented glucosamine to place itself into the tissues. It’s snake oil.

Very good you pointed this out as Stanfield superficially and carelessly trashes supplements doing a lot of harm to people who could otherwise benefit from them. He lacks critical indepth analysis needed to make an accurate judgement. I too have read that it makes the difference if it is the G-sulfate form since the cartilage is made up of glycoaminoglycans in sulfated forms, so if you lack the sulfate it seems to be an incomplete form for cartilage synthesis. Glucosamine sulfate stimulates the production of glycosaminoglycans (GAGs) in cartilage. GAGs are important building blocks of connective tissue, including cartilage, and are necessary for healthy joint structures. Glucosamine sulfate also inhibits the degradation of GAGs. Stanfield needs to go back to his biochemistry text books and look a little deeper.

I have several issues with Brad's summary. 1) Although NMN supplements do seem to raise blood levels, this only seems to happen consistently for doses above a half gram daily. Some of the experiments Brad mentions had a lower dose, so unsurprising they showed no effects. 2) NMN seems to allow older people like me to tolerate more exercise, to recover faster, and it is the increased exercise that seems to benefit us. I think the experiments Brad mentions looked for benefits from NMN without asking participants to exercise more and that may have masked the benefit of NMN. 3) Immune system improvements are vitally important as we age, seemed to happen in mouse experiments, but I don't think have been tested in human experiments yet. I seem to be much less likely to get sick when I am taking NMN. Yes, thanks Brad, I and every other literate human on the planet already know what the placebo effect is, so granted more testing is needed, but this seems very promising to me. 4) Most people are not taking NMN alone. Like many others, I take NMN, TMG and Apigenin together and do seem to experience a synergistic benefit. 5) I experienced one benefit that seems definitely due to NMN, because a problem I had experienced for years and which my mother had before me, disappeared within days after I increased my NMN dose from .25 gram to 1 gram daily... we had a tendency to get sleepy when driving and had to fight to stay awake on any drive over 20 minutes or so. This problem just disappeared. It doesn't happen to me any more.

I have always caught 3 to 4 colds every autumn/winter period, but since I started NMN, 0,5 go now 1 to 1,25 daily, I haven't had one in 4 years...

@@slimturnpike All personal experience is "anecdotal". However, we happen to be living in unusual times. Sometime in the next 10 or 20 years, for the first time in human history, extending life-span will become possible. For an older person, the rational analysis is " do nothing, resulting in certain death" or "study the existing limited research and play the odds, as best one can read them" and try to survive until the first breakthrough happens. Waiting passively is not an appealing option.

Absolutely spot on. Brad, as usual, continues his hate campaign against Sinclair, albeit more cautiously and subtly nowadays. I’m amused how he selects mainly negative research studies for his ‘talks’. I find his campaign interesting in so far as I see a display of open antagonism from a relatively literate man ( whatever that may mean). Sinclair had to block him eventually and left Brad whining about his t. Let’s hope he eventually gets what he himself has become.

That's why he is conservative with the dosing, compared to other supplements (hence "micro" vitamin). Ideally you want to set up your own stack based on your needs, and determining these precisely can be difficult. If you eat well and have no deficits, then maybe you don't need any supplements at all.

What’s your age? I feel like NMN will probably only have effects for people over a certain age. Like say growth hormone, where it’s so high when you’re younger, no need to boost it

@@kevinsutube1p528 like with many supplements, if you feel a difference then it's probably because you had a deficit in something. Personally I don't feel a difference with a lot of supplements that supposedly help people (and I'm close to 50), which I interpret as a good sign :)

Resveratrol did nothing for me while NMN provides visible results, both in terms of stamina and immunity. I'm 67 and play racket sports 3 to 4 times a week, Donotage NMN is my provider. Sinclair is trying to get NMN stricken off the list of food supplements by the FDA and classify it as a drug.

I’m 47 yrs old and started hot flushes early summer. I started doing my research on longevity. I started consuming soil milk and started taking Nmn last summer. My hot flushes disappeared in 1 less than a month of taking nmn and soy milk. My periods are so regular and my fogginess improved and my energy level increased. I’m a Nurse I work 12 hrs 4 days a week. My stamina improved and I still have thick dark long hair. I think NMn helped me a lot. I’m already almost taking 1 year of NMn from do not age. I also added taurine, speemidine and sirt 6 this January

I've read studies on all of those. The most promising appear to be glycine and taurine. I've tried supplementing with taurine (I'm past 60 so my level should be low) but my body doesn't accept it well. It made me quite dizzy. I tried lowering the dose (don't remember what it was but about 1/3 that recommended). It left me feeling chilled to point that although I felt ok, I lost motivation to do anything. In my case, it may be that I already have plenty of taurine as since COVID I've had mild fibromyalgia symptoms and that can mess up amino levels so I'm probably not typical. Glycine is a very promising supplement. It too declines with age and supplementation seems to provide a number of benefits. The main one for health being a significant increase in the production of glutathione, the body's most potent anti-oxidant. Glutathione also decreases with age and is critical for maintaining mitochondrial health. Glycine is also well tolerated in very high doses - much higher than you need to gain from its benefits. It can also increase creatine. I've been taking it for a while. I just add it to drinks, it tastes like sugar. I've seen the studies on NAC but the only good studies that seem to show benefits are those that combined it with glycine. Then studies using glycine alone seemed to give similar results to the combination. Also, as I understand it cysteine is not something that declines with age. I heard one explanation that it may help as a sulfur donor in the production of glutathione. In that case, if you eat cruciferous veg or eggs while taking glycine, that should have the same effect as well as being cheaper and safer. I eat plenty of both. Interesting to see how cause and effect have been misinterpreted in the case of age-related NAD+ decline. Rather than NAD+ decline leading to less energy expenditure, it's just the opposite!

Taurine and Glycine cancel each other out in liver uptake when taken together. Taurine can be sedating due to its effect on GABA, very noticable when taking 1 gram on empty stomach with glass of water.​@@antonystringfellow5152

Funny story re the placebo effect. Once I was walking through a park on a hot day, and I came across a woman who was nearly passed out in the middle of a path with a small crowd around her. In my experience people sometimes get lightheaded when they're out in the heat too long and haven't eaten enough, so I figured that maybe some sugar could help and gave her some mints that I had on me. She eats the mints and then gets up, says she's starting to feel better and leaves with some friends. A few minutes later something occurs to me, and I take a closer look at the the packaging that the mints came in. I then turn to my friend sort of surprised / sort of confused because those mints were sugarless...

Don't tell stories like that. As the father of a type 1 diabetic who intentionally keeps himself low and often overshoots the mark, I can tell you that the need for glucose is absolutely real, the effects are almost immediate and no amount of placebo effect can bring a low diabetic back to normal.

They likely just recovered and stood back up and the person above is the only one who concluded that the ill person needed sugar from mints. Maybe she just felt better just because of the short rest and attention/aid from another person.

@anna-xz9yj Of course. That person was clearly not diabetic. You know that, and I know that. But I don't want people to think that the person was diabetic (obvious conclusion from the story) and that severely low blood glucose isn't real. Having to recently revive my son from severe low glucose, I can tell you it was a harrowing experience. No amount of fake glucose could produce a placebo recovery.

This video is interesting because he doesn’t lay out the actual facts and then proceeds to promote his vitamins. That sounds like a pharmaceutical rep. Taking 1000 mg of NMN is not the same as taking 1000 mg of niacin (Vitamin B3), and it does not equate to an extremely high dose of Vitamin B3 in the same way. The body tightly regulates the conversion of NMN to nicotinamide. This regulation helps prevent the buildup of excessive nicotinamide, reducing the risk of potential toxicity associated with high levels of niacin.

I felt ZERO difference. Absolutely nothing. In fact I stopped and improved my sleep a bit and feel fantastic off it. Million other things to do first before wasting money on this. Sleep exercise will do 10x more than NMN

I took it for 3 months and felt no difference, but also took blood tests before and after those 3 months. No difference there as well.

Ditto on that, I took it for a couple years, no noticeable difference also. Stopped altogether unless there's new evidence. My antiaging choice is Carbon 60 and recently Urolithin A (juries out on that one).

Same here. I seem pretty immune to placebo effects, and it's hard to find anything that makes a difference for me. So I'll stick with sensible eating, exercise and sleep.

There are two kinds of people who might benefit from NMN: 1) those with suboptimal NAD from any number of reasons 2) any sort of athlete who might potentially get better energy performance Studies testing a vitamin in people with optimal levels makes no sense.

I also took it for a year from a reliable source, DoNotAge. I never noticed any effects. I am 62 and in pretty good shape. There is always a risk of negative side effects, so I stopped taking it.

If you do adequate cardio exercise weekly and eat a good diet and get enough sleep, you're in a nad+ levels will be adequate, there won't be any benefit. People under 30 years of age have good NAD+ levels even if they don't do lots of cardioI NMn is a lifeline for me as I recover from a major health issue at age 48. Makes a huge difference in my energy, the health of my gums, and many other things I noticed from taking it. I just don't get the same performance from niacin. The other trick that I've found is that much higher doses are effective taken in small doses throughout the day. I take at least three to four grams a day and sometimes up to 6 if I don't get enough sleep. But there's only works if I divide it in small doses that I take every few hours. I think taking large doses at once ends up being eliminated from the body before these doses can be utilized.

Dr. Stanfield is a doctor of medicine and is trained to diagnose and treat diseases. I’m sure he’s great in his profession but he’s not a Doctor of Human Biology and his opinion is no different than the regular Joe. It’s two completely different fields. Find a Doctor of human biology who focus on the underlying biology and chemistry of the human body without the clinical focus on disease management. I once heard a discussion about eating real food vs. eating the stuff we produce today that "looks like food" but has a lot of things removed. The statement was something along the line of, "we're eating something that LOOKS like a vegetable, but it's missing all these trace vitamins and minerals that exist for a million different reasons." The point was, we don't know why vitamins, minerals, and antioxidants all exist in the miniscule quantities they exist in nature... and there's probably a damn good set of reasons why they exist that way (which is a roundabout way of saying we evolved alongside nature and this is just "how it works"). So now days we offer these artificially created, "singled out" vitamins, minerals, antioxidants, and various other supplements ... but they aren't alongside the trace vitamins, minerals, and antioxidants that they are found alongside in NATURE. What does that mean? How many TRILLIONS of chemical reactions happen in our bodies as we process these vitamins, minerals, and antioxidants? How many pathways are unlocked by a trace amount of this chemical or that molecule when we consume a veggie that has a key vitamin we don't seem to ABSORB and UTILIZE when we take a supplement instead? TRILLIONS of reactions across the TRILLIONS of microbes and cells in our bodies every day. Literally not even possible to measure these accurately with a quantum computer. It's THAT COMPLICATED. And yet, it's so simply if we eat organic foods that are as close to nature as possible. Trillions of reactions every day and nature already has it all figured out (with OBVIOUS exceptions, nothing is perfect 100% of the time). All we know for sure... it's a hell of a lot more perfect when we stay close to nature than when we mess with it and try to artificially replicate it!!

The neurological benefits of raising NAD (such as with NR) will be what is important. By the way, why did the sibling mice that were given NMN look so much younger than their sisters and brothers? In my case, my mild psoriasis was cured, and I do not feel stiff in the mornings anymore, and I know people with lupus, and neuropathy who greatly benefit. A young person or mouse who eats well and exercises will not benefit from increasing NAD with supplements. Their NAD levels would be high already, and cells would not seek an additional supplement.

Placebo or not, glucosamine and chondroitin relieves my knee pain. When i dont take it my knees hurt. When i do take it my knees feel much better. Period. .. its been that way for well over 20 years. If it wasnt for glucosamine and chondroitin i would not be able to walk now.

I think the point of NMN is to increase capacity for cell repair and activation of good genes. That does not imply that it cures arterial diseases or reverses glucose etc. I think it is proven that NMN activated sirt genes which relates to longevity, but I may be wrong. I recommend to take TMG as well if you take NMN to make sure that you have enough methyl groups, it can be important.

I would take him more seriously if he didn't spend so much time bashing Sinclair. Also, isn't intracellular NAD levels the most important measure, not blood levels? Get that measured before and after. I experimented with just niacin and my NAD intracellular was 44. Then started taking 1g of nmn again and feel better. Need to redo my test now.

The body is actually proving time after time, that the body can heal itself and or adapt to what the mind believes about the body. Great work and a thorough explanation. I really liked the bottom line. NMN may prolong the life of mice because it may protect them from cancer. But NMN doesn't protect against the cardiovascular disease and therefore most likely does not affect the life expectancy of humans.

If you want nad precursors to work you need to fix nad salvage and other pathways of nad dysfunction. If the NAD you supply rapidly converts to NADh then it could be adding fuel to metabolic disorder fire. A long post like this will probably be shadowbanned but.. My controversial analysis shows that excessive NAD donors dysregulated certain forms of catecholamines. That plasma norepinephrine, epineprine, normetanephrine levels become dysregulated with excess NAD precursor supplementation. Some of which can be good or bad. Its a weird melting pot of data that is hard to decipher.

A study published in Nature Medicine, February 2024, a strong correlation was shown between niacin metabolites (esp. PY-4) and cardiovascular disease. This raises concern about using niacin, nicotinamide, NR, and NMN supplements. The study was done by a noted cardiologist at the Cleveland Clinic. Comments?

I took on a regular basis 2-3gr of NMN per day for 18 months and I wanted and almost believed to have results... but when it was time to renew my NMN reserves I made a test by stopping completely just to see if my sports performances decreased... No change... NMN is expensive, I spent 1200$usd in 18 months without any improvement whatsoever. To this day, I don't know of any product that really improves my performance... I'm hopeful with glycine, which I've started taking at a rate of 12-18gr a day... I've tried so many things in 4 years... if you have any ideas, let me know.

I tried NMN and it often gave me diarrhea and didn't really make me feel any better (possibly worse?). I think because it's bitter it makes you feel like you're getting an energy boost. Having said that, I am a person that can often get diarrhea if I drink too much coffee and bad foods, so it could be just me. The thing that has made me feel the best is running everyday. Exercise is king. Don't run out and buy supplements when you start feeling sick and tired. Just maintain your health with eating well and exercising, it's way cheaper in the long run.

My experience is with NR (300 mg). NR supplement does appear to be most available to the liver. To that end, my very high NAFLD markers came back down to within the reference range. Additionally, I had been receiving cortisone shots in my knees like clockwork every 100 days for several years. Within weeks of starting NR, not only have I not needed a cortisone shot in over 3 years, I have not experienced one single pain in my knees! Not a placebo effect as I had zero expectation that NR would have any effect on my knees. NR was the only new thing introduced (Rx or supplement) during that time. Presumably this was due to possible anti-inflammatory effects.

The final point you raised is concerning, however as I understand it, since NMN is a derivative of B3, it may not directly increase metabolites 4PY & 2PY like other forms of B3. I always wondered why some companies that sell supliments, particularly elite level sports supliments, like "monster pack" multivitamins add so much vitamins, particularly from the B family, sometimes going up to 20.000% of the daily dose. anything that increases skeletal muscle - that usually means weight training. We lose muscle as we age, aerobic activities are beneficial but don't build or maintain enough muscle to offset the effect of sarcopenia. Substitute some of your 50 chins (overkill) for lower body weight bearing (lunges, squats, deadlifts) and add in some overhead presses. You need to challenge your muscles, and at 50 chins, your muscles are not challenged. Add weight. Lots of it. Your reps will go down but your strength go up, which is apparently the predictor of health/longevity.

I have been taking nmn for a year... I felt more energetic overall, but I have been taking other supplements like spermidine, taurine, fisetin, and so on. I eat one meal a day. Every week I stop eating for 48 hours once. Every month I do not eat for 72 hours once. I do all this because I drive long haul trucks and I sit for about 10 hours a day do not have the time to exercise. I have not had any side effects until recently I had nerve pain as if someone was pushing needles on my leg, left arm and middle chest. It happens rarely but it does last an hour pain comes and goes.

Actually I’ve been experimenting with NMN and “possibly” it worsened all of my aging markers on my withings scales over the past month, im still trying to figure out which of my supplements did it because i did several changes, so now im trying to single it out, but i suspect it was NMN, plus i have been struggling to keep my body fat levels low - that might correlate to the insulin sensitivity you discussed.

What I'm wondering is how can a supplement be made that directly effects muscular NAD levels. While exercise does increase NAD levels in muscle, the amount of which it increases still diminishes with age. I think NMN is on the right track but I'm hoping further r&d will be done in order to produce a new form that can directly increase NAD levels in tissue. Being able to keep muscle NAD levels from falling as you age would be a huge deal.

Well... just because NMN is decreased doesn't necessarily mean increasing it would improve health outcomes. For example, cholesterol drops as you get older, due to reduced consumption of food, yet we KNOW reducing blood APOB (part blood cholesterol) is a good idea.

@@adamc2320 Incidence of dry eyes with finasteride is incredibly low. And Dr Stanfield himself stated that it’s related to tretinoin in the past.

I’ve heard they’re mostly rancid so I switched to straight up cod liver oil. I do Nordic naturals too just as a bonus but keep it in the freezer. I’d never use the ancient grody ones from cheap sources after watching that report a few years ago about rancid pills.

That's only a small problem for people with A-fib and taking very high doses of fish oil in the 3-4g+ EPA/DHA. They're fine at 1g EPA/DHA

Nice video - correction, body converts NMN to nicotinamide(niacinamide) not niacin.
