Immunohistochemistry (IHC) techniques involve the use of antibodies to detect specific proteins or antigens within tissues. The process involves the binding of primary antibodies to the target antigen followed by a secondary antibody conjugated with a detectable marker such as a fluorescent dye or enzyme. This allows for the visualization and localization of the target protein within the tissue sample. An immunohistochemistry test involves the application of IHC techniques to detect specific proteins or antigens in tissue samples. This can be used to identify specific cell types, diagnose diseases, and monitor therapeutic responses. To perform immunohistochemistry, tissue samples are first fixed and sectioned onto slides. These slides are then treated to remove endogenous peroxidases and block non-specific binding sites. Primary antibodies specific to the target antigen are then applied to the slides, followed by secondary antibodies conjugated with a detectable marker. The slides are then washed and counterstained before being viewed under a microscope. The components identified by immunohistochemistry can vary depending on the specific target antigen being studied. This can include proteins, hormones, enzymes, and other biomolecules. Immunohistochemistry is used for a variety of applications, including the identification of specific cell types, the diagnosis of diseases, the monitoring of therapeutic responses, and the evaluation of research hypotheses. IHC stands for immunohistochemistry.

I have a question regarding colorimetric detection: for the secondary antibody to work, in the cell there must be a process that is regulated by the enzyme attached to the secondary antibody, right? And does this process have to do with the original protein that interests us, the one that is recognized by the primary antibody? Thank you

Please do not confuse between immunofluorescence and immunohistochemistry? What happened to antigen retrieval? this video can misguide people

Exactly what I was thinking. He kept mixing up the steps between IF and IHC. IHC needs antigen retrieval step done with a buffer and exposed to heat and pressure, not triton or other detergent like you mentioned for permeabilization which is done for IF.

These terms are really subjective , when you use enzyme based dete tion it's histochemistry, when you use fluorescence based detection it's immunofluoresence...but the principle is same.
