When Whitebeard said he told Ace to go, he meant as his father he watched him leave when he knew he could have stopped Ace from leaving by force if needed. He's just taking responsibility as a parent.

The story definitely emphasizes inherited will having more importance than blood ties. As you said, Luffy is a lot like Roger and Ace like Whitebeard, even if they're not blood related. Always saw it as when Whitebeard said he told Ace to go, it was him trying to relieve some of Ace's guilt about the whole war situation.
