Saponification value is defined as the amount of potassium hydroxide (KOH) in milligrams required to saponify one gram of fat or oil. It is a very important testing parameter for oil or fat samples. This video represents a complete procedure for the determination of Saponification Value of an oil or fat sample.

when i did this experiment, i mixed 3.4g KOH+ 2ml distilled water+rest of ethanol to make upto 100 ml volume(0.5 N solution). I made this ethanolic KOH using directions I found on the web. Then I directly dissolved 1g oil sample in 50 ml of prepared mixture. It got clear only in 5 minutes. I prepared 0.5 N Hcl by adding 10ml in 250 ml water(37% Hcl). When I titrated, I was getting a very large saponification value(about 500). The formula I used is [(blank titre-sample titre)*56*normailty of KOH]/weight of sample. Can you tell me what am I doing wrong? And what is the purpose of the methods u have used like use of aluminium foil and initially boiling koh with ethanol?

Great video, can you explain what is function of reflux 1,67g KOH in 200mL absolute ethanol? Can we only use 6g KOH in 150mL absolute ethanol for preparing 4% ethanolic KOH? Thanks..

Reflux of ethanol with KOH followed by distillation is normally done to make the ethanol more active for this test. But you can just dissolve 6g in 150ml.

When refluxing or heating the KOH and fat...the KOH disappears leaving something like a whitish powder. I then need to refill again with KOH. Why does this happen?

in the standard formula for SV, it uses the molecular weight of KOH (56.1g/mol), but in this method, it applies 28. 05? my question is what this number represents?

Mike Tyson, which standard formula are you talking about. In this test, we followed the AOAC 920.160 official method strictly. In AOAC 920.160 method, 28.05 is used in the equation, so we did.

@@MicroChemsExperiments This is because HCl concentration is 0.5 N. Basically the equation involved HCl concentration too. When it is 0.5 N Molecular wt of KOH is 56.1 which will eventually lead to the value of 28.05.

We need to dissolve KOH in cold environment to prevent the vigorous reaction of KOH with ethanol which may lead to the loss of properties of KOH solution.

Sample flask contains sample, thats why during saponification, the concentration of KOH in sample is decreased, thus HCl is needed more in compared with blank.

Yes, because molecular weight decreases after hydrolysis & as the mol wt decreases SV in increase,so definitely SV of split fatty acid will be more than oil before hydrolysis.

Both calculations are same. You will get the same result. Second equation, you mentioned, is simplified form of calculation. First mentioned calculation is the original calculation formula. You can use both.

Martin Okyere, Thanks for your observations. You are right about error. We should use 100ml burette to avoid refilling. We have tried to refill it fast to avoid the error as we dont have 100ml burette.

@Goutam Gupta, this test method and calculation are strictly followed the AOAC 920.160 official method. AOAC given the calculation. The reason behind it is: Sample and blank are Titrated with the same concentration of HCl (0.5N), thats why 0.5 become minimized from each side of sample and blank.

@@tarunkumar8841 Sorry, it was a mistake. 28.05 is the correct factor. So 28.05 should be written in second line also. But we wrote 28.5 by mistake.

It is a half of KOH molecular weight 56.11%2 = 28.05

@@amanueldaniel5342 Just dissolve KOH pellets in absolute ethanol and skip the distillation step. Take help from this saponification video and just skip the distillation part and disoolve KOH in ethanol

You can prepare 0.5N HCl solution by followng our video: kzfaq.info/get/bejne/or6qdKVp3NWllGw.html Just multiply the amount of HCl, needed to prepare 0.1N HCl, by 5. Then follow the whole procedure to standardize the solution.

@@factxfact5549 To test an oil quality, you must go for the following tests: Peroxide value, iodine value, Free Fatty Acid and Acid Value, Rencidity, Saponification value, Unsaponifiable matter, RM value, Polensky Value, Moisture, Refrective Index, Density etc.
