The joke is that, now everyone wants Jesus to turn water into wine, whenever they are given the chance to ask him, since turning water into wine is one of the things he is famous for doing. This is all hypothetical and for the purpose of a joke though. The other Jesus joke was that Jesus said that he is the way and that all who follow him will know the way (I guess in life? But I'm not sure). So, when the Uber driver asks Jesus "do you know the way (to your destination)?" Jesus responds with "Yes, I am the way."

And I find the jokes fall a little flat. I mean I guess there's a little comedic value in the modernization of something into an uber situation but for the most part I feel like the video is meant to make people laugh to demonstrate that they understood it rather than because it was a really cleaver take. There were a couple that were worth a chuckle, like the water into wine situation though even that doesn't fit the context well as it couldn't be legally consumed while driving which is the whole setup.
