My anecdotal story: I'm a 60 year old male in Palm Springs, CA. I started taking NR(NAD precursor) about 6 months ago. I dropped from 240 pounds to 190 pounds and have kept the weight down, but bounce up and down with about an 5 pound variation. I believe the benefit of NR was to stimulate my exercise program which is basically 1 hr walk in the morning, 1 hr walk in the evening, plus intermittent gym based light weight lifting on machines. I have also tried variants of fasting, but have found that a 5 day water fast consuming only water and green tea works extremely well from a psychology point of view. Once I get past the first 24 hr fast, then the next 4 days are much easier because my appetite is suppressed. I'm taking NR during the fast. Also, started watching Dr. Stanfield about the same time I started the NR regimen(began with 300 mg per day, but now take 600 mg per day). The youtube support from Brad and others is invaluable. Thank you for helping everyone.

Not sure how green tea fits into all that fasting. EGCG has been shown to increase senescence. Not what something one should aim for if one is over 60 .

Finally, a definitive interview about resveratrol. For me, its time to close the chapter on it. Also, there was always something wrong about Dr Sinclair's impish smile, like he knew something that we don't, so now, we know it too.

See, this is great. We need more videos like this. I think that my presence is not appreciated on several KZfaq channels because I question David Sinclair's veracity and contributions to the field of anti-ageing. I know he likes attention and publicity, and that he is making A LOT of money at something. (Lab experimentation doesn't pay that well.) I have read through hundreds of papers (I am a chemist, but involved in art restoration) and do not find evidence for the life extending protocols that Sinclair promotes.

​@@Pluvo2for1 Right now I usually refer people to the work of Valter Longo. He's a solid and sincere researcher who doesn't make outrageous claims. Periodic fasts or employing fast mimicking (Prolon or water fasting) for 5 days about 3x a year looks promising. A plant-based Mediterranean-type diet comes out at the top for good health, limiting protein intake and employing moderate IF (12-14 hours daily) to reduce mTOR, moderate alcohol consumption (1-2 glasses of red wine,) and moderate daily exercise - these are your best bets right now and dovetails with rapamycin research which looks promising. Also, do not look to purchase rapamycin - this needs to be researched more. Taking rapamycin unregulated can be dangerous. Make sure you get enough vitamin D + K and try to de-stress. Dr. Brad is doing some good work here. I'm impressed (and that's saying a lot. I've never been impressed with David Sinclair.) Feel free to ask any questions, Pluvo2for1. 🌹

Thanks Dr Brad and Dr Matt. I also learn that we have to honestly find out what works and what doesn't, when it works and when it doesn't. Be happy to know when there's a good finding that disproves our beliefs. Our beliefs are not the reality. The map is not the territory. It's very important to do reality check consistently in any discipline, and advices.

Peter Attia and others have been talkin about rapalogs for years now I think. 57:04 "Nobody has shown yet that you can do partial reprogramming in an old mouse and actually restore youthful function...", except David Sinclair's lab did a year ago.....the study was titled "Reprogramming to recover youthful epigenetic information and restore vision" and it was published in Nature. The lead author was Yuancheng Lu. How could he have missed that? "...or increase lifespan" - In order to do this you would have to deliver the gene therapy to an old mouse's failing internal organs. The concept is pretty sound, however, the required biological engineering to accomplish it remains to be worked out. That is why Sinclair's lab started with the eye, because it is a complex organ that can be accessed without cutting open the animal. I have to wonder, for a guy whose next big study is the effect of intermittent fasting on canines, why he feels the need to throw so much shade at people's work.

Thanks for the great video! It would also be interesting to learn what people can do now. Especially when you are 70 or 80 years old and don't have so much time to wait for well-founded study results and established therapies... of course every year that you gain can decide if you reach age escape velocity … The question for me is always: what is applicable without major risks and of course no "snakeoil"

I have an anecdotal feeling that my very low biological age is mainly due to High Intensity Resistance Training 3-4 times a week over 45 years. I think it ultimately protects the immune system, stimulates hypertrophy (age 70 at 70kg and bench 100kg) and improves lactic acid utilisation through the cori cycle...

Being too sceptical is as useless as not being sceptical at all. He spent most of the interview negating pretty much everything, and the little research information he did have, he decided to withhold anyway. Don't forget about the power of placebo Mr Kaeberlein.

Great interview… I’ve been taking 1g NMN per day for 6 months with 30g olive oil. No discernible difference in resting HR; HR variability; sleep quality; body weight or FTP (cycling)… Still taking it though because I’m a sucker for an expensive placaebo

Fascinating and exciting interview! I was very surprised to hear that some people don’t or might not respond well to caloric restriction based on their genetics. I haven’t found any discussions about this online yet…I thought caloric restriction was nearly universal for most/all organisms for either extending lifespan or improving health. Can you or anyone in the comments section here clarify?

1:15:37 An interesting comparison would be Rapamycin verses Metforman vs. 1-3 days of fasting. It could be that fasting provides the most benefit with AMPK activation (and with no side affects).

Nice video. A small question: Does it make sense on restday afternoons when i take berberin, to reduce my leucin rich proteins, to not interfere with mtor reducing effect of berberin? Or wouldnt it really matter?

I had a similar experience with a rotator cuff tendinopathy. After 9 months of pain, two cortisone shots and a couple of months of physical therapy it was still inflamed and not healing. I'm a biologist, so I hit the books on tendinopathy. Then I took fisetin for 4 days to kill senescent inflammatory cells and followed up by stacking collegen peptides, AKG and vitamin C (to build new collagen) and curcumin (to repress residual inflammation). The tendinopathy disappeared in less than three weeks and in two weeks more I had my range of motion back.

@@Pakistan-Icecream It was an integrated systems level approach - fisetin to stop the collagen destructive senescent SASP and the collagen stack to help remodel the damage. If I had to guess, I'd say fisetin was the more important, but the rest were synergetic.

Glycans are a really interesting measure of biological ageing. In addition, it would be interesting to cross check with methylation. I'm in the process of checking this out on myself. I'll report back when I get the results.

I am 50 and take 3mg Rapamycin (amongst other things, including NMN) on a Monday and lift weights (split routine) on Thursday, Friday, and Saturday.

He didn't specifically address fasting as a way to block mTOR aside from just calorie restriction. Do we have any data showing that fasting blocks MTOR as well as rapamycin? I'm thinking that his once a week rapamycin dose could be mimicked by weekend fasting? Any thoughts?

Does rapamycin restore mtor sensitivity similar to keto for insulin resistance ? We always assume more is better but i feel the right amount at the right time and sufficient sensitivity would be wiser? Insulin is great but clearly we overuse it. Perhaps same for mtor?

18:01 1. feeling mentally sharp is very different from actually being mentally sharp. you feel sharp on lower carb, bc of increased stress hormone concentrations (impaired mental ability across the board compared to moderate to high carb) 2. feeling physically sharp is very different from actually being physically sharp (low carb decreases work capacity, relative perceived intensity for the same intensity increase on low carb, motor unit recruitment decrease on low/lower carb ,which in turn lowers the potency of any working set done in the gym for a given movement) ......
