Totally agree about happiness being a factor! My grandma is 96, lives alone without any need for help and the only med she is on is blood- lowering medication and only every other day, because taking it every day makes her dizzy. She is the most optimistic and happy person I have ever met and I am pretty sure that goes a long way!

Here's my two cents' worth, being an extremely muscular, fit, and healthy guy in his late forties: 1) Exercise, especially add weight training four times a week. 2) Get your diet right, reduce simple carbs, increase protein and soluble fibre, add hemp seed oil (or hemp seeds) to at least one meal daily, since it contains the perfect balance of Omegas 3, 6, and 9. I add hemp seed oil and hemp seeds to my daily leafy salad or parsley salad. 3) Stay hungry. Don't ever overeat or feel excessively full. 4) Drink black coffee, with no sugar, during the day, and try to eat your first meal later in the day. 5) Sleep well. 6) Stay mentally and socially engaged, challenging your mind daily. 7) After all that, then you can add supplements like D3&K2, niacin, magnesium, zinc, creatine, hyrdolyzed collagen peptides, SAMe, NAC, turmeric, berberine etc. But the supplements are the final piece of the puzzle. Anyway, from my experience and my perfect health and extremely muscular body, this has worked wonders for me.

Thank you for this solid and down-to-earth interview, Dr. Stanfield. However, the two main strategies suggested for healthspan/lifespan - exercise and calorie restriction - conflict each other, since regardless of whether we do cardio or resistance training, we will always need increased calorie intake, if we don’t want to start losing muscle. This being said, I think many of us would appreciate it if you find a way to give us your feedback on this issue. Thanks again!

well my placebo reaction to NMN is far superior to any anti-depressant I've tried in the last 10 years and I'm keeping it. I can now get through a workout without crashing and after a month on it I now feel ready to live a normal life; I wasn't functioning before I started it. I did not expect such a response. Don't like the insomnia that goes with it but in terms of getting me to function daily, I now need it, don't want to be without it and if they make it unavailable, I'm going to be lost.

I love the way Prof Kaeberlein presented his views and ideas on all these supplements because this has been my view point as well. Very unbiased and calls out the error in the system with how supplements are branded without the need for any real substance behind them. My rule as a scientist is, if i cant understand the biological mechanisms of how a substance acts and the litterature data is conflicting, then take it with a huge grain of salt because we know labs will push for a certain outcome just to satisfy the people funding the research.

Prof Kaeberlein is the most skeptical scientist I've heard in this field, and that's a good thing. He's the exact opposite of Dr Sinclair, who was the elephant in the room during this conversation.

55:10 - Appreciate this point by Prof. Kaeberlein that simply telling one 'you should take this or that for longevity' is irresponsible or simplistic for a highly complex subject. Actually, "the more uncertain (leaving more room) one's saying the more scientific/correct he is". Decisions one has to make at some point are a compromise of pro and con. 1:01:43- well, that is only practically speaking. Scientifically tho, being able to live long in unhappiness would be very interesting for understanding longevity. Actually there is no sci proof of 'happiness' benefits on longevity. People just say so - sci or fi ? If it does, then next happiness must be clearly defined for different kinds. 1:22:51- No, peptides are hydrolized to AAs then absorbed, in small intestine, well before large intestine.

Collagen is not meant to be absorbed in whole. It can't. Rather, It is its AA profile that matches that of the target tissues, skin, tendon,... so that the absorbed AAs originated from the collagen supplement are utilized to synthesize these tissue's proteins at high efficiency due to the matching AA profile. Its effects can be literally seen of faster fingernail growth when one takes the supplement.

I'm glad you presented the info for collagen peptides Dr. Brad. I do hope they work. I do the hyaluronic, collagen peptide powder and a proretinol cream. Spf in the morning or before sunlight. I do think it helps. Thank you for following the research closely.

This was a great podcast. Matt is giving a very realistic view of where science is with longevity. Not like dr Sinclair… only think I’d say is I do think zone 2 is important from personal experience. It’s not very hard to do or figure out how to do it and the injury risk is very low.

For me, it's not just been all the money dished out for these supplements, but the time spent researching them. You start taking a supplement, then find more information on it where people are saying "well, if you take supplement A, you must also take B to counteract C that might be caused by A". Before you know it you're taking 3 extra supplements because they supposedly work synergistically or fix each other's side effects. Then all those supplements in turn seem to have riders; a particular supplement has to be the kind that's excreted by polar bears and filtered through a platinum strainer or it won't be effective. You want to take a fish oil supplement? Better research all the brands' oxidation reports. Hyaluronic acid? Some say get high molecular weight because the low one causes inflammation, others say to get the low molecular weight because the high one won't penetrate. Now I read I'm supposed to look for mid-molecular weight, and good luck finding out from the manufacturers how many kilodaltons their product is. Then once you pick one and spend all that money, someone will come out with a video or blog on why that supplement isn't effective at all or why they stopped taking it (no offense meant, Dr.) and why this new study shows it. For people who love to dive deep into this subject and love the science and experimenting I'm sure that's not a big problem, but for some there just comes a point where they go: "F*ck it, I'll just eat my veggies and hope for the best."

this video shows longevity research is going at a SNAIL's pace. Very very sllllloooooowwww. Cant even tell if RAP will cause loss of muscle mass. maybe 3-5 yrs from now Just barely learning how to test more than one compound at a time. Good Lord, we should be able to test several at a time. That might come in 300 years from now.

The muscular system is a highly metabolic and adaptable system that secrete a wide a array of substances. Avoiding/reducing sarcopenia might be helpful in many more ways than just to keep people’s strength at a level that make them functionally independent and make their daily life manageable. (Conflict of interest - I take CreatineAKg and creatinemonohydrate)

Push back a little when he says things that are wrong. The collagen peptide meta analysis was on clinical trials, not epidemiological studies. Would have also been nice to ask about a few of the following issues: 1. Evidence that Rapamycin can increase amyloid beta in a particular AD model (he generally dismisses this unfavorable data on Rapa AFAIK - exactly an issue that he criticized other scientist of) 2. Rapa generally causes weight loss in mice. I would presume it does so by reducing caloric intake. This seems to confound the Rapamycin data in general (is it partially just a drug that induces CR). Any concerns for using rapamycin for men for reproductive health? (recent findings for metformin had some birth defect issues!). Agree with skepticism on supplements. Only thing I take if EPA/DHA since it seems to have the most evidence compared to all other supplements.

Love the content as usual Dr. Brad this one was truly an eye opener for me. As I've always suspected, nobody really knows how different humans respond to supplementation. I've also wondered about multiple supplementation it could be highly detrimental to health and we just don't have the data to prove otherwise. On this basis I may cut my supplement stack considerably. FYI no mention of Ca-AKG is this because of lack of data??

He kept saying caloric restriction is the best intervention but what does that mean? Caloric restriction of 1 calorie below BMR? 10% of BMR? What is an effective calorie restriction? Would appreciate an answer or a steer

Really bright guy and quite conservative concerning supplements. We have all known for years that some of the data on supplements is questionable at best. Nevertheless, we also know for sure what happens if we take no supplements at all, we get old, feeble, etc and often live for many years in this condition. So to me the downside being the cost of the supplement is worth the it even if it improves my life just a little bit. I still take vit d, creatine, a multi, circumin, and focus heavily on a plant based diet and regular exercise.

Regarding collagen studies, I’m confused about his (Prof Kaberlein) use of the word epidemiological when Dr. Brad was referring to placebo controlled clinical trials. Those are not epidemiological studies. I originally had the same questions that Professor Kaberlein did regarding the breakdown of collagen protein in the gut into individual amino acids. But there is evidence to show they get absorbed as di and tripeptides.

I am diabetic and metformin has caused extreme muscle weakness. If muscles are a sink for glucose disposal, how does metformin make sense when just walking becomes an insurmountable challenge? I have taken myself off metformin and the difference in my muscle strength is like night and day. Walking, exercise even running is doable. So what is more dangerous not taking metformin or not maintaining lean muscle & strength? I fear staying on metformin will result in being wheelchair bound then agecare. Metformin makes me miserable so what is the solution? I am happier being off meds because I can be active and strong.

why rapamycin over the other mtor inhibitors? also rapamycin also inhibits mtorc2, causes a lot of side effects like lung toxicity and diabetes symptoms, and increased risk of certain cancers... who will take this knowing these side effects?!

100% agree. Suppresses your innate immune system and side effects evident quickly even at low weekly anti-aging dosing. Who want lower lymphocytes and compromised natural immune system?

iirc Sinclair said that massive induction of DNA breaks without causing mutations leads to accelerated epigenetic aging in models and consequent display of aging phenotype. That does seem highly suggestive that epigenetic aging is behind the aging phenotype.

The most complicated way to manage aging entirely ? Manipulate matter cell by cell, molecule by molecule. The simpler / easier ones are epigenetic manipulations for which current research is just looking into, let alone providing a medical application at this moment in time. A lot will be learned and modeled, many of these interventions will leave imperfections as they address some bigger factors. Overall we should be able to reach more thorough procedures to effectively deal with aging entirely within 30-50 years given the progress of computational technology and the current renewed interest in biotech and aging. Then perhaps 70 year in the future (30 years later) we can use all that data and understanding as well as forms of nanotechnology to provide "perfect" age controlling interventions.

At 55:45 you mentioned creatine and you've talked about it on other videos. Well, it may work for some people, but I've tried it for an extended trial twice because you've been enthusiastic about it. For me, I've noticed no effect whatsoever either good or bad. I do exercise with moderate intensity some days and high intensity on alternate days, but I am over age 70. So, maybe that has something to do with it. The tests I used are the total number of pullups I can do (8 sets of 10) and the 100 yard dash (4 sets). I noticed no improvements with these two tests. Any thoughts?

P M After my hip operation, I started creatine. I believe it got me fit and healthy very quickly with use of weights and kettlebells etc. When I ran out, the urge to do weights had gone. After further research, I understand you have to load creatine, 20grams for each of 7 days, after that about 5gram each day. So I started yesterday, around 20gms and will continue for full 7 days, so until next Monday, after that I shall take 3 to 6 grams each day. So within 3 weeks, I should see an improvement. I understand it is very quick result after loading is done.

By the way I have just started pull ups again in the last week or so and not getting very far. I am hoping to see vast difference in my pull ups and the time to increase to 5 minutes of practice instead of 30secs.

Certain long lived species of bats experience substantial epigenetic rejuvenation each hibernation cycle, this may or may not be influential towards their great longevity. I tend to suspect it might be highly influential but I could be wrong. We definitely need to see epigenetic reprogramming tests in mice.

also, if any of you struggle with migraines or frequent head aches. the primary cause is the shortage of vitamin b1, b2 due to its short half life in the body and magnesium, but magnesium has a long half life and doesn't need a frequent supplementation. the best form is the bio-coenzymated forms of the b1 and 2 as these are fat soluble and easily cross the brain blood barrier. also, if you are physically active, supplementation of collagen will prevent any excess joint wear, which is common among elderly due to the poor production and athletes due to excess consumption leading to rapid premature wear. in women soy based Phosphatidylcholine (PC) along with a mild use of vitamin k2 will prevent any formation of diseases associated with blood vessel plaque. such as a stroke, dementia, alzheimers and many neuro-degenerative diseases. also the occasional use of methyl folate (not folic acid) a vitamin b9, vitamin b12 and b6 will further stabilize the cns and the brain function in the long term. as well as promote faster healing/detoxification associated with strong methylation. with this formula you can easily make it up to 110 with nearly zero visits to any doctors whatsoever, which be annoying/costly/unnecessary. what more can you possibly want.

Interesting study on mice by Satchin Panda with evidence of circadian eating being a relevant factor not due to diet restriction. Both mice feed same caloric western diet only changing feeding time and those with restrictive feeding time lost weight. Not sure if this has been done on humans😢

The key here is that not all protein is the same. Proteins are very, very different depending on what types you're talking about. Fish is best, and legumes. Im not an official medical professional, but I know, that if we want to increase longevity, we need to be able to change our perspective and therefore our cultural programming, which will give us lots of new data. Atmospheres, contamination and surfaces are extremely important, but still most doctors dont take those things into account. Also Kundalini will help us to understand our body much better, which also will give us more data. Also we can have objects in our eyes, which will make our whole body sick and those are extremely hard to remove again, bc they are deep inside and bc the eye is so soft. I think no doctor really understands it, right now. If we remove those, we def will live longer and better imv.

When my Cat was sick she stopped eating for 3 days and it seems there is an instinct to restrict food for autophagy/ longevity. Anyway on day 3 I gave oral anti biotics because she has a flu, shes doing well now and has a good appetite.

It seems to work best in older mice. Also, it lowers the immune system, and can cause diabetes. Not a drug that is ready for prime time...

Yes, this is known and should be put into practise. Older people need more protein. I'd start at age 50.

Plant based protein doesn't have the same negative effect on the body as animal derived. There is a lot of science showing this. Go to pubmed and look.

Data re Interventions on already healthy middle-age people are the only ones that matter to 90% of your audience. Sleep, eat well, exercise, socialize in person THEN let’s see what these add-ons can do. Eye health, agility, ability to solve problems one has the skills to solve….

It would ,its an AMPK antagonist so all exercise will be muted. Not completely but you will notice gains aren't as much as before. AMPK activates when we exercise so if your already taking Metformin and exercising your AMPK won't react when you exercise, at least as intensely as it does without the Metformin and your gains won't be as much.
